17β-雌二醇和维生素K2对人脐带间充质干细胞成骨分化的影响
Effect of 17β-estradiol and Vitamin K2 on Osteogenic Differentiation of Human Umbilical Cord Mesenchymal Stem Cells
【摘要】 目的 探讨17β-雌二醇和维生素K2对人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,h UC-MSCs)成骨分化的影响。方法 选取2019年11月内蒙古妇幼保健院的1份脐带标本,分离培养出h UC-MSCs。选择10-10 mol/L17β-雌二醇(17β-estradiol,E2)、10-7 mol/L维生素K2(vitamin K2,VK2),将细胞分为4组,分别为E2组、VK2组、E2+VK2组、对照组。实时荧光定量聚合酶链式反应检测目的基因骨基因Runt相关转录因子2(recombinant runt related transcription factor 2,RUNX2)、Ⅰ型胶原(type I collagen,COL1)、碱性磷酸酶(alkaline phosphatase,ALP)的表达。结果 成功提取出h UC-MSCs,通过茜素红、油红O、甲苯胺蓝染色证实h UC-MSCs具有多向分化的能力。结果显示与对照组相比,E2或VK2促进COL1(0.853、0.847)、RUNX2(0.830、0.757)、ALP(0.127、0.137)的表达,且联合组(COL1:1.003、RUNX2:1.170、ALP:0.163)作用更明显,差异有统计学意义(t=24.022、56.541、18.500;15.119、48.833、34.617;25.000、38.000、41.000,P<0.05)。结论 E2和VK2能够促进h UC-MSCs成骨分化,且联合作用效果更好。本研究为治疗骨损伤性疾病提供新的思路,同时为临床应用提供理论基础。
【Abstract】 Objective To investigate the effects of 17β-estradiol and vitamin K2 on osteogenic differentiation of human umbilical cord mesenchymal stem cells (h UC-MSCs).Methods A sample of umbilical cord was taken from Inner Mongolia Maternal and Child Health Care Hospital in November 2019,and h UC-MSCs were isolated and cultured.Select 10-10 mol/L 17 β-estradiol (E2),10-7 mol/L vitamin K2(VK2),the cells were divided into four experimental groups,adding E2 group,VK2 group,E2+VK2 group,control group.The expressions of target genes recombinant runt related transcription factor 2,RUNX2,type I collagen,COL1 and alkaline phosphatase,ALP were detected by real-time polymerase chain reaction.Results HUC-MSCs were successfully extracted,and the multi-direction differentiation ability of h UC-MSCs was confirmed by alizarin red,oil red O and toluidine blue staining.The results showed that compared with the control group,E2 or VK2 promoted the expression of COL1(0.853,0.847),RUNX2(0.830,0.757) and ALP(0.127,0.137).The effect of the combined group (COL1:1.003,RUNX2:1.170,ALP:0.163) was more obvious,and the,and the difference was statistically significant (t=24.022,56.541,18.500;15.119,48.833,34.617;25.000,38.000,41.000,P<0.05).Conclusion E2 and VK2 can promote the osteogenic differentiation of h UC-MSCs,and the combined effect is better.This study provides a new idea for the treatment of bone injury diseases,and provides a theoretical basis for clinical application.
