高湿度环境下犬脓皮病病原葡萄球菌感染小鼠的试验研究
Experiment on infection of mice with Staphylococcus spp. from canine pyoderma under high humidity
【摘要】 为了研究犬脓皮病病原菌的致病机制、筛选有效的临床外用药物,采用砂纸打磨皮肤的方法,选取临床犬脓皮病病例中分离得到的伪中间葡萄球菌(Staphylococcus pseudintermedius)和金黄色葡萄球菌(Staphylococcus aureus)各1株,进行分点皮内注射感染小鼠,并于室内常规湿度(30%~40%相对湿度)和高湿度(60%~65%相对湿度)环境下饲养96 h,记录临床表现、剖检变化及病理组织学观察。结果显示:常规湿度下感染小鼠均于2~3 d结痂康复,而在高湿度环境下造模小鼠均出现明显的临床症状。病理切片结果显示,在高湿度环境下,2种菌感染小鼠的皮肤各层及皮下组织病理变化与临床上犬脓皮病病例相似,伪中间葡萄球菌感染组小鼠皮肤多出现脓肿灶,可见明显囊膜结构;而金黄色葡萄球菌感染组多为弥散性变化。结果表明,60%~65%相对湿度环境有利于犬脓皮病病原葡萄球菌感染小鼠皮肤。
【Abstract】 In order to lay a foundation for the study of the pathogenic mechanism of canine sepsis and the screening of topical drugs, this experiment was conducted using the clinical canine sepsis pathogen Staphylococcus spp. to infect the skin of mice. One strain of Staphylococcus pseudointermediate and one strain of Staphylococcus aureus were isolated from clinical cases of canine pyoderma respectively. Intradermal injections of the strains were performed at points after sanding the skin with sandpaper, and the infected mice kept for 96 h under normal indoor humidity(30%-40% relative humidity) and high humidity(60%-65% relative humidity). We recorded the clinical manifestations, observed the pathological slices by necropsy, and found that the mice infected under the normal humidity recovered from the scabs on 2-3 days of the experiment, while the mice modeled under the high humidity environment all got positive clinical symptoms. The results of pathological sections showed that in the environment of high humidity, the pathological changes in the skin layers and subcutaneous tissue of the mice modelled with the two strains were similar to the clinical cases of canine pyoderma. In the S. pseudointermediate infected group, ulcers were formed on the skin; abscesses could be seen under the ulcers, and the capsule structure was obvious and visible, while the S. aureus infected group mostly presented diffuse diseases. These results indicated that the 60%-65% relative humidity environment would be conducive to the infection of mouse skin by Staphylococcus from canine pyoderma.
