糖尿病大脑葡萄糖代谢紊乱介导认知功能障碍的研究进展
Review of brain glucose metabolism disorder promoting cognitive dysfunction in diabetes mellitus
【摘要】 糖尿病(diabetes mellitus, DM)是最常见的全身性代谢性疾病之一,可损害机体多个器官,已成为全球范围内的重大公共卫生问题。90%~95%DM病例被归为2型糖尿病(type 2 diabetes mellitus, T2DM),约60%~70%的糖尿病患者存在轻度或中度认知功能障碍,糖尿病引起认知功能障碍与大脑葡萄糖代谢紊乱密切相关。大脑葡萄糖代谢紊乱可通过能量不足、蛋白质O-Glc NAc糖基化减少和氧化应激等机制引起大脑神经元受损。本文就T2DM中大脑葡萄糖代谢紊乱的病因和导致认知功能障碍的机制进行综述,为糖尿病认知功能障碍的防治提供一定参考。
【Abstract】 Diabetes mellitus(DM) is one of the most common metabolic diseases, which can damage multiple organs and has become a major public health problem worldwide. Among the confirmed DM patients, type 2 diabetes mellitus(T2DM)accounts for 90% to 95%, and about 60% to 70% of diabetic patients show mild or moderate cognitive impairment. The cognitive impairment caused by diabetes is closely related to the disorder of brain glucose metabolism. Neuron damage can be induced by glucose metabolism disorders through the mechanisms of energy deficiency, reduced protein O-GlcNAc glycosylation, and oxidative stress. The etiology of cerebral glucose metabolism disorder in T2DM and the mechanism leading to cognitive dysfunction were reviewed in this article, to provide reference for the prevention and treatment of cognitive dysfunction in diabetes.
