中华猕猴桃根提取物增敏索拉非尼对H22肝癌小鼠移植瘤生长及TNF-α的影响

【摘要】 目的 观察中华猕猴桃根提取物增敏索拉非尼对H22肝癌小鼠移植瘤生长及肿瘤坏死因子-α(TNF-α)的影响。方法 将24只H22肝癌造模成功的小鼠按随机数字表分为4组:肝癌组给予生理盐水灌胃,猕猴桃根提取物组给予中华猕猴桃根提取物灌胃,索拉非尼组给予索拉非尼灌胃,联合组给予中华猕猴桃根提取物和索拉非尼灌胃,均1次/d,连续灌胃15 d。荧光素酶标记观察各组小鼠移植瘤生长情况,计算各组小鼠出瘤时间、瘤重、抑瘤率,免疫组化SABC染色法观察瘤组织中TNF-α表达情况。结果 索拉非尼组、联合组小鼠出瘤时间均明显长于肝癌组(P均<0.05),瘤重均明显低于肝癌组和猕猴桃根提取物组(P均<0.05),抑瘤率均明显高于猕猴桃根提取物组(P均<0.05);联合组小鼠出瘤时间明显长于索拉非尼组(P<0.05),瘤重明显低于索拉非尼组(P<0.05),抑瘤率明显高于索拉非尼组(P<0.05)。肝癌组瘤组织中TNF-α表达增多,猕猴桃根提取物组、索拉非尼组、联合组TNF-α阳性表达率均明显低于肝癌组(P均<0.05),索拉非尼组、联合组均明显低于猕猴桃根提取物组(P均<0.05)。结论 中华猕猴桃根提取物可能通过下调炎症因子TNF-α的表达增敏索拉非尼对H22肝癌小鼠的抑瘤作用。

【Abstract】 Objective It is to observe the effect of Actinidia Chinensis root extract sensitizes sorafenib on transplanted tumor growth and tumor necrosis factor-α(TNF-α) in H22 hepatocellular carcinoma mice. Methods Twenty four mice successfully modeled H22 liver cancer were divided into 4 groups according to the random number table: the liver cancer group was given normal saline by gavage, the Actinidia Chinensis root extract group was given Actinidia Chinensis root extract by gavage, the sorafenib group was given sorafenib by gavage, and the combined group was given Actinidia Chinensis root extract and sorafenib by gavage, all once a day, continuously treated for 15 days. The growth of transplanted tumor in the mice of each group was observed by luciferase labeling, the time of tumor emergence, tumor weight and tumor inhibition rate of each group were calculated, and the expression of TNF-α in tumor tissues was observed by immunohistochemical SABC staining. Results The tumor emergence time of the mice in the sorafenib group and the combination group was significantly longer than that in the liver cancer group(both P<0.05), the tumor weight was significantly lower than that in the liver cancer group and Actinidia Chinensis root extract group(both P<0.05), and the tumor inhibition rate was significantly higher than that in the Actinidia Chinensis root extract group(both P<0.05); The tumor emergence time of the combined group was significantly longer than that of the sorafenib group(P<0.05), the tumor weight was significantly lower than that of the sorafenib group(P<0.05), and the tumor inhibition rate was significantly higher than that of the sorafenib group(P<0.05). The expression of TNF-α were increased in tumor tissues in the liver cancer group, the positive expression rates of TNF-α in the Actinidia Chinensis root extract group, sorafenib group and combination group were significantly lower than that of liver cancer group(all P<0.05), the rates in the sorafenib group and combined group were significantly lower than that in the Actinidia Chinensis root extract group(both P<0.05). Conclusion Actinidia Chinensis root extract may sensitize the anti-tumor effect of sorafenib in H22 hepatocellular carcinoma mice maybe by down-regulating the expression of inflammatory factor TNF-α.
