新型溴化阻燃剂TBPH对雄性SD大鼠免疫毒性研究
Immunotoxicity of TBPH,a novel brominated flame retardant, in male rats
【摘要】 目的 探讨2,3,4,5-四溴-苯二羧酸双(2-乙基己基)酯[bis(2-ethylhexyl) tetrabromophthalate, TBPH]对雄性SD大鼠免疫功能及免疫细胞比例的影响。方法 48只雄性SD大鼠随机分为4组,每组12只,分别为对照组(玉米油)和5、50、500 mg/kg·bw TBPH剂量组,灌胃28 d后使用流式细胞术测定大鼠外周血、脾、胸腺及肠系膜淋巴结的免疫细胞比例;用刀豆蛋白A和细菌脂多糖诱导淋巴细胞转化实验测定脾淋巴细胞增殖能力;用乳酸脱氢酶法检测NK细胞活性;用荧光胶珠法测定腹腔巨噬细胞吞噬能力。结果 各剂量下未观察到大鼠的体重和相对脏器重量有改变。免疫细胞分型的结果显示,与对照组相比,50 mg/kg·bw(LSD-t=2.50,P<0.05)和500 mg/kg·bw(LSD-t=2.45,P<0.05)组大鼠脾T细胞比例降低,差异有统计学意义。淋巴细胞增殖能力测定实验,观察到50 mg/kg·bw(LSD-t=2.50,P<0.05)和500 mg/kg·bw(LSD-t=3.09,P<0.05)的TBPH可降低T淋巴细胞的增殖能力,差异有统计学意义,B淋巴细胞增殖能力无改变。未观察到NK细胞活性和腹腔巨噬细胞吞噬能力有改变。结论 经口暴露50和500 mg/kg·bw的TBPH可抑制雄性大鼠脾T淋巴细胞的增殖能力,降低脾T淋巴细胞的比例,对适应性免疫有一定的影响。
【Abstract】 Objective To assess the effects of bis(2-ethylhexyl) tetrabromophthalate(TBPH) on immune function and the proportion of immune cells. Methods Fourty-eight male SD rats were randomly divided into control(corn oil), 5, 50, and 500 mg/kg·bw TBPH-treated groups(n=12 per group). All rats were gavaged once a day for 28 consecutive days. Flow cytometry provided the information about proportion of primary immune cells from peripheral blood, spleen, thymus, and mesenteric lymph nodes. The proliferative capacity of T and B cell from spleen was detected by MTT assay after concanavalin A(Con A) and lipopolysaccharide(LPS) stimulation. The killing activity of NK cells from spleen was assessed by lactate dehydrogenase assay. Fluorescent latex beads were used to examine the phagocytosis of peritoneal macrophages. Results No significant changes in body weight and organ coefficients were observed in the rats within the dosages of this study. Compared with the control group, the proportions of T cells in spleen was decreased by TBPH at dose of 50(LSD-t=2.50, P<0.05) and 500 mg/kg·bw(LSD-t=2.45, P<0.05). Meanwhile the proliferation of T cells separated from spleen were suppressed in the rats who were treated with 50(LSD-t=2.50, P<0.05) and 500 mg/kg·bw TBPH(LSD-t=3.09, P<0.05). At the same time, there were no significant changes in the proliferation of B cells, the killing activity of NK cells, and the phagocytosis of peritoneal macrophages. Conclusion Oral exposure to 50 and 500 mg/kg·bw TBPH would inhibit the proliferation of splenic T lymphocytes and decrease the proportion of splenic T cells in male SD rats.
