电针对AD模型大鼠梨状皮质GalR2和AchE表达的影响
Effects of electroacupuncture on the expression of GalR2 and AchE in piriform cortex of AD rats
【摘要】 目的:观察电针(EA)对阿尔茨海默病(AD)模型大鼠梨状皮质甘丙肽受体2(GalR2)和乙酰胆碱酯酶(AchE)表达的影响,旨在探讨GalR2及AchE在电针治疗AD过程中的相互作用。方法:将雄性SD大鼠40只随机分为5组:对照组(C组)、模型组(M组)、EA+M1145组、EA组、EA+M871组。双侧海马内微量注射Aβ1-42,每侧5μL进行造模。EA组予以电针“大椎”“百会”“足三里”穴每天30 min,连续4周。EA+M1145组和EA+M871组分别经侧脑室注射M1145和M871后进行电针干预。水迷宫检测大鼠学习行为能力,组织学染色和Western blot法检测GalR2和AchE的表达。结果:M组平均逃避潜伏期延长(P<0.05),梨状皮质的细胞多呈空泡状,GalR2和AchE的表达均增强(P<0.05)。EA后,大鼠平均逃避潜伏期缩短(P<0.05),细胞形态趋于正常,GalR2的表达进一步增加(P<0.05),而AchE的表达降低(P<0.05),两者的变化以EA+M1145组最为明显(P<0.05)。结论:EA可能通过调节GalR2和AchE的表达改善AD模型大鼠的学习记忆能力,并且GalR2的高表达对AchE有抑制作用。
【Abstract】 Objective:To observe the effect of electroacupuncture(EA) on the expression of galanin receptor 2(GalR2) and acetylcholinesterase(AchE) in piriform cortex of rats with Alzheimer disease(AD) for clarifying the interaction of GalR2 and AchE in the treatment of AD with EA.Methods:Forty male SD rats were randomly divided into control group(C group),model group(M group),EA+M1145 group, EA group and EA+M871 group.Aβ1-42 was injected into bilateral hippocampus in micro-dose( 5μL/side) to establish the models.Rats in EA group were treated with EA at the points of "Dazhui"(DU 14), "Baihui"(DU 20) and "Zusanli"(ST 36),30 min a day, for 4 consecutive weeks, and those in EA + M1145 group and EA + M871 group were injected with M1145 and M871 into lateral ventricle, respectively, followed by EA intervention as the protocol described in EA group.Morris Water Maze was used to detect the learning and behavior ability of rats.Finally, histological staining and Western blot were used to observe the expression of GalR2 and AchE in piriform cortex of the rats.Results:Average escape latency was significantly increased in M group(P<0.05).The cells at piriform cortex of rats in this group mostly presented in vacuole shape, with enhanced expression of GalR2 and AchE(P<0.05).After EA,the average escape latency was shortened(P<0.05),and the cell morphology tended to be normal.GalR2 expression was further increased(P<0.05),yet AchE expression was down-regulated(P<0.05).The changes of GalR2 and AchE were dominant in EA+M1145 group(P<0.05).Conclusion:Electroacupuncture may improve the learning and memory ability of AD model rats by regulating the expression of GalR2 and AchE,and higher expression of GalR2 could inhibit AchE expression.
