鲜黄连体外酶抑制活性及稳定性研究
Study on Enzymatic Inhibitory Activity and Stability of Plagiorhegma Dubium. in Vitro
【摘要】 目的:测定鲜黄连3种不同溶剂提取物中9类化学成分的含量、体外酶抑制活性,并进行了相关性分析,考察了不同条件下鲜黄连提取物的稳定性.方法:采用紫外分光光度法测定鲜黄连的甲醇、乙醇和水3种提取物中9类化学成分含量,体外评估其对α-葡萄糖苷酶、α-淀粉酶、乙酰胆碱酯酶、丁酰胆碱酯酶、黄嘌呤氧化酶、脲酶和酪氨酸酶的抑制活性,并基于上述结果对活性最佳的甲醇提取物进行了体外胃肠道、pH和热稳定性的研究.结果:鲜黄连甲醇提取物的总糖、总酚、总黄酮、总鞣质、缩合鞣质和没食子鞣质含量较高,鲜黄连乙醇提取物的总三萜、总酚酸含量较高,水提取物的总蛋白含量较高.鲜黄连甲醇提取物对α-淀粉酶、乙酰胆碱酯酶和丁酰胆碱酯酶抑制活性较强,其最高活性分别为47.1±2.3 mg AE/g extract、19.6±1.6 mg DE/g extract、2.5±0.1 mg DE/g extract.相关性分析显示,9种化学成分均与体外的酶抑制活性呈一定相关性(r=-0.98~1.00);确定鲜黄连甲醇提取物为最佳,其在3种稳定性研究过程中具有良好的表现.结论 鲜黄连9种化学成分和体外酶活性具有较高的相关性,并在不同条件下相对稳定,为扩大天然药用资源提供了研究方向.
【Abstract】 objective To determine the contents of 9 series of chemical components in different three solvent extracts of Plagiorhegma dubium. Methods The contents of 9 series of chemical components in methanol,ethanol and aqueous extracts of Plagiorhegma dubium were determined by UV spectrophotometry. Meanwhile,the inhibitory activities of α-glucosidase,α-amylase,acetylcholinesterase,butyrylcholinesterase,xanthine oxidase,urease and tyrosinase were determined in vitro. Results The methanol extract of Plagiorhegma dubium had high contents of total carbohydrate,total phenol,total flavonoid,total tannin,condensed tannin and gallotannin,the total triterpenoid and phenolic acid were higher in the ethanol extract,the content of total protein was higher in the aqueous extract. The methanol extract of Plagiorhegma dubium had strong inhibitory activities on α-amylase,acetylcholinesterase and butyrylcholinesterase. The results of correlation analysis showed that the 9 series of chemical components were correlated with the enzymatic inhibitory activity in vitro(r=-0.98~1.00). Conclusions The 9 series of chemical components of Plagiorhegma dubium have high correlation with the enzymatic activities in vitro,and are relatively stable under different conditions,which could provide research directions for expanding natural medicinal resources.
