基于16S rRNA测序技术探究参苓白术药膳糊对功能性腹泻脾虚证大鼠肠道菌群的影响
Investigation of the effect of Ginseng and Atractylodes medicinal paste on the intestinal flora of rats with functional diarrhea due to spleen deficiency based on 16S rRNA sequencing technology
【摘要】 目的 观察参苓白术药膳糊对功能性腹泻脾虚证大鼠肠道菌群的影响,探讨参苓白术药膳糊治疗功能性腹泻脾虚证的机制。方法 将30只SPF级Wistar大鼠按随机数字表法分别为正常组、模型组、蒙脱石散组、参苓白术汤剂组、参苓白术药膳糊组,每组6只,正常组大鼠正常饲养,余鼠以高乳糖联合水平台站立方法制作功能性腹泻脾虚证大鼠模型。造模成功后,对应给药治疗7d。治疗结束后采用16S rRNA基因测序技术分析大鼠粪便菌群变化。结果 造模后正常组大鼠体重显著重于其他组(P<0.05)且造模大鼠腹泻率为100%。治疗7d后,参苓白术药膳糊组大鼠体重显著重于其他造模组(P<0.05)。在门水平,参苓白术药膳糊可以显著增加厚壁菌门、变形菌门、放线菌门丰度,降低疣微菌门、拟杆菌门丰度;在属水平,参苓白术药膳糊可以增加乳杆菌属、异杆菌属、双歧杆菌属,降低拟杆菌属、瘤胃球菌属、阿克曼菌属、螺杆菌属、颤螺菌属、副拟杆菌。结论 参苓白术药膳糊可以改善功能性腹泻脾虚证,其作用机制可能与增加肠道中有益菌丰度,调节肠道菌群结构有关。
【Abstract】 Objective To investigate the effect of Ginseng and Atractylodes macrocephala on the intestinal flora of rats with functional diarrhea with spleen deficiency, and to explore the mechanism of Ginseng and Atractylodes macrocephala in treating functional diarrhea with spleen deficiency.Methods Thirty SPF grade Wistar rats were divided into normal group, model group, montmorillonite group, ginseng and atractylodesis soup group and ginseng and atractylodesis medicinal paste group according to the random number table method, six rats in each group. After successful modelling, the rats were treated for 7 d. After treatment, the rats were analysed for changes in faecal flora using 16S rRNA sequencing technology.Results The rats in the normal group weighed significantly more than the other groups(P<0.05) and the diarrhea rate of the rats was 100%. After 7 d of treatment, the rats in the Ginseng and Atractylodes group weighed significantly more than the other groups(P<0.05). At the phylum level, Ginseng and Atractylodis Macrocephalae Paste could significantly increase the abundance of Phylum Thick Bacillus, Phylum Aspergillus and Phylum Actinomycetes, and decrease the abundance of Phylum Wartybacter and Phylum Bacillus. At the genus level, Ginseng and Atractylodis Macrocephalae Paste could increase the abundance of Lactobacillus, Isobacillus and Bifidobacterium, and decrease the abundance of Bacillus, Rumex, Ackermannia, Helicobacter, Treponemae and Paramecium. Conclusion The mechanism of action may be related to increasing the abundance of beneficial bacteria in the intestinal tract and regulating the structure of intestinal flora.
