针刺对CRS抑郁大鼠海马糖皮质激素基因表达的影响与相关基因生物信息学分析

【摘要】 目的 针刺治疗慢性束缚(chronic restraint stress, CRS)抑郁模型大鼠,观察大鼠海马糖皮质激素基因的变化,利用生物信息学分析针刺后逆转基因与神经精神疾病的相关性。方法 将雄性SD大鼠分为三组:对照组(9只),束缚组(9只)和针刺组(9只)。适应性喂养后进行CRS造模,造模后针刺组进行治疗。行为学采用旷野实验(open field test, OFT)、强迫游泳实验(forced swimming test, FST)和新环境抑制实验(novelty suppressed feeding test, NSFT)。实验结束后取三组大鼠海马进行基因检测与生物信息学分析。结果 造模后大鼠表现出运动活动减弱、记忆受损、焦虑样和抑郁样行为,针刺治疗可以逆转上述行为。糖皮质激素基因检测结果显示:对照组大鼠和束缚组有19个差异基因。针刺治疗后,束缚模型大鼠以上19个差异基因中有11个被逆转。生物信息学分析表明,被逆转基因在双相情感障碍、焦虑症、精神分裂症、阿尔茨海默病和抑郁症的相关基因组中高度富集。结论 针刺能够逆转束缚模型大鼠行为学和糖皮质激素相关基因的表达,生物信息学证实逆转的基因与神经精神疾病相关基因组密切相关。补充针刺治疗抑郁症糖皮质激素基因水平的相关机制。

【Abstract】 Objective To observe the changes of glucocorticoid gene in hippocampus of chronic restraint stress(CRS) depression model rats treating by acupuncture, and analyze the correlation between the reversal gene after acupuncture and neuropsychiatric diseases using bioinformatics. Methods Male SD rats were divided into three groups: control group(n=9), CRS group(n=9) and acupuncture group(n=9). After adaptive feeding, CRS model was established, and acupuncture group was treated after model establishment. Behaviors adopted open field test(OFT), forced swimming test(FST) and new suppressed feeding test(NSFT). After the experiment, the hippocampus of the three groups of rats was taken for gene detection and bioinformatics analysis. Results After modeling, the rats showed decreased motor activity, impaired memory, anxiety and depression like behaviors, and acupuncture treatment could reverse the above behaviors. The results of glucocorticoid gene detection showed that there were 19 different genes in the control group and the restraint group. After acupuncture treatment, 11 of the above 19 differential genes in the CRS model rats were reversed. Bioinformatics analysis shows that the reverse gene is highly enriched in the genomes related to bipolar disorder, anxiety disorder, schizophrenia, Alzheimer’s disease and depression. Conclusion Acupuncture can reverse behavioral and the expression of glucocorticoid related genes in CRS model rats. Bioinformatics confirms that the reversed genes are closely related to the relevant genomes of neuropsychiatric diseases. Mechanism of glucocorticoid gene level in depression treated with acupuncture supplement.
