少阴病(心肾阳虚证)“欲解时”PGC-1α通路蛋白表达的节律变化及四逆汤干预作用的研究

【摘要】 目的 通过检测心肾阳虚证模型大鼠在“欲解时”及其对应时段心肌组织能量代谢通路关键蛋白的变化,探讨少阴病“欲解时”的时间治疗学意义。方法 SD大鼠随机分为欲解时正常组、非欲解时正常组、欲解时模型组、非欲解时模型组、欲解时药物组、非欲解时药物组。造模及给药结束后,分别于少阴病“欲解时”及“非欲解时”取心肌组织,连续6d。Western Blot检测欲解时和非欲解时实验大鼠心肌组织PGC-1α、NRF-1、TFAM蛋白水平。结果 模型组和药物组PGC-1α、NRF-1、TFAM的蛋白表达大多为欲解时高于非欲解时;药物组心肾阳虚证大鼠的PGC-1α、NRF-1、TFAM通路蛋白表达量较模型组高。结论 欲解时PGC-1α、NRF-1、TFAM通路的蛋白表达量较非欲解时高。四逆汤对心肾阳虚证模型大鼠PGC-1α、NRF-1、TFAM通路蛋白的表达有上调作用。

【Abstract】 Objective In this study, this study explored the temporal therapeutic significance of ShaoYin disease(cardiac-kidney-yang deficiency) by using the changes in key proteins expressions of key proteins of the energy metabolism pathway of myocardial tissue during the "Yujie time" and corresponding periods in the model of cardiac-kidney-yang deficiency. Methods SD rats are randomly divided into normal group at "Yujie time", normal group at "Non-Yujie Time", model group at "Yujie Time", model group at "Non-Yujie Time", drug group at "Yujie Time", and drug group at "Non-Yujie Time". After the end of molding and administration, the myocardial tissue was taken from “Yujie Time” and "Non-Yujie Time " for 6 days. Western Blot detected the levels of PGC-1α, NRF-1, TFAM protein in experimental rat myocardial tissue during the Yujie Time and Non-Yujie Time.Results The expression of proteins in PGC-1α, NRF-1 and TFAM in the model group and drug group was higher than that in Non-Yujie Time, and the expression of PGC-1α, NRF-1 and TFAM pathway proteins in rats with cardiovascular and renal yang deficiency in the drug group was higher than that in the model group.Conclusion The expression of protein in PGC-1α, NRF-1 and TFAM pathways was higher in Yujie Time than Non-Yujie Time. The Sini Decoction had an upregulating effect on the expression of PGC-1α, NRF-1 and TFAM pathway proteins in rats with cardiac-kidney yang deficiency model.
