沉默S100P基因抑制肺腺癌细胞A549增殖、迁移和侵袭
S100P gene silencing inhibits proliferation, migration and invasion of lung adenocarcinoma A549 cells
【摘要】 目的:探讨肺癌组织及细胞中S100P基因表达对增殖、迁移、侵袭的影响及其作用机制。方法:在GEPIA(Gene Expression Profiling Interactive Analysis)数据库和HPA数据库(Human Protein Atlas)得到肺癌数据,分析S100P基因在不同分型肺癌组织与正常组织中差异表达。使用shRNA-S100P RNA下调肺癌细胞系中S100P基因表达水平;细胞的增殖活性使用MTS比色法检测;细胞周期变化运用流式细胞仪检测;细胞划痕实验和Transwell实验分别检测细胞迁移能力和侵袭能力。RT-qPCR与Western blot分别验证S100P表达及细胞周期、凋亡蛋白表达水平。结果:S100P基因在肺癌组织中表达水平明显高于正常组织(P<0.01);不同分型的肺癌组织中S100P基因表达存在差异。与对照组相比,下调S100P使A549增殖、迁移和侵袭能力均降低。结论:S100P基因在肺癌中高表达且下调后可抑制A549细胞的增殖、迁移和侵袭能力。
【Abstract】 Objective:To investigate the effect of S100P gene expression on proliferation, migration and invasion in lung cancer tissues and cells and its mechanism.Methods:The data of lung cancer were obtained from GEPIA(Gene Expression Profiling Interactive Analysis) database and HPA(Human Protein Atlas) database, and the S100P gene was analyzed for the differential expression of S100P gene in different types of lung cancer tissues and normal tissues.The expression of S100P gene in lung cancer cell line was down regulated by shRNA-S100P RNA.The cell proliferation activity was detected by MTS colorimetry.Cell cycle changes were detected by flow cytometry.Cell scratch test and Transwell test were used to detect the migration ability and invasion ability of cells respectively.RT-qPCR and Western blot were used to verify S100P expression, cell cycle and apoptotic protein expression.Results:The expression level of S100P gene in lung cancer tissues was significantly higher than that in normal tissues(P<0.01).S100P gene expression was different in different types of lung cancer.Compared with the control group, down-regulation of S100P reduced the proliferation, migration and invasion of A549.Conclusion:S100P gene overexpression and down-regulation in lung cancer can inhibit the proliferation, migration and invasion of A549 cells.
