铁死亡相关蛋白SLC7A11及GPX4在肾细胞癌中的表达及意义
Expression and significance of ferroptosis related proteins SLC7A11 and GPX4 in renal cell carcinoma
【摘要】 目的:探讨SLC7A11及GPX4在肾细胞癌及正常肾组织中的表达差异,初步分析SLC7A11及GPX4的表达情况与肾癌预后之间的关系。方法:采用免疫组化方法检测SLC7A11及GPX4在肾癌及正常肾组织中的表达,同时分析SLC7A11及GPX4表达水平与肾癌患者临床病理参数的相关性。采用Spearman法分析SLC7A11及GPX4之间表达的相关性,利用Kaplan-Meier生存曲线及Cox回归方法分析SLC7A11,GPX4表达的高低及临床因素对肾癌患者预后产生的影响。结果:SLC7A11及GPX4在肾癌中为高表达,而在正常肾组织为低表达(P<0.05),SLC7A11及GPX4蛋白表达与肾癌大小、远处转移及临床分期密切相关,SLC7A11与GPX4之间呈正相关(r=0.583,P<0.01)。在肾癌组织中,SLC7A11,GPX4表达阳性的患者与阴性表达的患者相比预后较差(P<0.05)。多因素分析提示肾癌大小、远处转移、临床分期、SLC7A11及GPX4是影响肾癌预后的独立因素。结论:SLC7A11及GPX4在肾癌中高表达,可能参与了肾癌的发生,SLC7A11或GPX4高表达的肾癌患者预后不良,未来可能成为肾癌治疗新靶点。
【Abstract】 Objective:To investigate the differences in the expression of SLC7A11 and GPX4 in renal cell carcinoma and normal renal tissues, and to preliminarily analyze the relationship between the expression of SLC7A11 and GPX4 and the prognosis of renal cell carcinoma.Methods:The expression level of SLC7A11 and GPX4 in renal cell carcinoma and normal renal tissues were detected by immunohistochemistry, the correlation between the level of SLC7A11 and GPX4 and clinicopathological parameters of patients was analyzed.Spearman method was used to analyze the correlation between SLC7A11 and GPX4 expression, Kaplan-Meier survival curve and Cox regression were used to analyze the influence of the level of SLC7A11,GPX4 expression and clinical-pathological features on the prognosis of renal carcinoma patients.Results:SLC7A11 and GPX4 were highly expressed in renal cell carcinoma, while, were low expressed in normal renal tissue(P<0.05).In addition, the expression of SLC7A11 and GPX4 were closely related to tumor diameter, distant metastasis and the clinical stage of renal cell carcinoma.Further analysis showed that SLC7A11 and GPX4 were positively correlated(r=0.583,P<0.01).In renal cell carcinoma tissues, patients with positive SLC7A11 and GPX4 expression had a poorer prognosis than those with negative expression(P<0.05).Multivariate analysis indicated that tumor diameter, distant metastasis, clinical stage, SLC7A11 and GPX4 were independent prognosis factors.Conclusion:SLC7A11 and GPX4 are highly expressed in renal cell carcinoma, which may be involved in the occurrence of renal cell carcinoma.Patients with renal cell carcinoma with high SLC7A11 and GPX4 expression have a poor prognosis.
