不同诱孢方法对辣椒疫霉菌孢子囊消长动态的影响
Effects of Different Sporulation Methods on the Growth and Decline Dynamics of Sporangia of Phytophthora Capsici
【摘要】 为明确不同诱孢方法对辣椒疫霉菌孢子囊消长动态的影响,为以辣椒疫霉菌孢子囊为病原接种体的相关研究提供技术支撑,试验通过选取5种培养基并结合抹伤处理方式诱导孢子囊产生,研究在不同诱孢条件下辣椒疫霉菌孢子囊从产生到完全释放的变化动态差异。结果表明,5种供试培养基中,辣椒疫霉菌仅在PA、CA和V8培养基上可以产生孢子囊,孢子囊产生最大量分别为7.238万、3.712万、0.523万个/mL;OA和PDA培养基上未产生孢子囊。抹伤处理后,PA、CA和V8这3种培养基上最大产孢子囊量均增加,孢子囊产生最大量分别为8.412万、3.916万、0.669万个/mL,孢子囊产生时间提前,最大产孢续存期增加;其中PA培养基经过抹伤处理对辣椒疫霉菌孢子囊产生的诱导效果最佳,不仅产生的孢子囊量最多,而且抹伤处理后比未处理的培养基孢子囊产生时间提前了2 d,最大产孢续存期增加了2 d,孢子囊的释放时间增加4 d。此外,OA培养基上也有孢子囊产生,但所需的时间较长,PDA培养基上仍未产生孢子囊。
【Abstract】 In order to find out the influence of different sporulation methods on the growth and decline dynamics of sporangia of Phytophthora capsica to provide technical support for related research on the sporangia of Phytophthora capsici as inoculum, in this study, five kinds of culture media were selected to induce sporangium production combining with smearing treatments, and then the dynamic difference of sporangia from production to complete release under different sporulation conditions was studied. The results showed that sporangia were produced only on PA, CA, and V8 media among the five tested media, and the maximum sporangium production was 7.238×104, 3.712×104, 0.523×104 sporangia/mL, respectively. No sporangium was produced on OA and PDA medium. After smearing treatment, the maximum sporangium production on PA,CA, and V8 media was increased, and the maximum sporangium production was 8.412×104 sporangia/mL, 3.916×104sporangia/mL, and 0.669×104 sporangia/mL, respectively. The production time of sporangium was advanced, and the maximum sporangium duration was increased. Among them, PA medium with smearing treatment had the best induction effect on the production of sporangia of Phytophthora capsica. The quantity of sporangia was not only the most on PA medium, but also the production time of sporangia was advanced by 2 d, the duration of maximum sporulation was increased by 2 d and the release time of sporangia was increased by 4 d. Besides, sporangia were also produced on OA medium, but it took a long time.Sporangia were still not produced on PDA medium.
