半夏软腐病拮抗细菌的筛选与鉴定

【摘要】 为解决半夏软腐病带来的种植弊端及经济损失,采集了湖北钟祥地区半夏种植合作社的健康半夏根际土,并从中筛选到2株具有较强拮抗半夏软腐病的病原菌胡萝卜软腐果胶杆菌性能的菌株JK-4和JK-28。根据其形态特征、生理生化特性并结合16S rRNA基因序列分析,将这2株菌鉴定为枯草芽孢杆菌(Bacillus subtilis),并将其分别命名为B.subtilis JK-4(JK-4)和B.subtilis JK-28(JK-28)。研究发现:JK-4和JK-28均具备较强的产脂肽类抗生素surfactin的能力,在高糖Landy培养基中,经30℃、200 r/min培养48 h后,surfactin的产量分别为1.28和1.12 g/L,且同时具备产NH3、分泌铁载体、产ACC脱氨酶及吲哚乙酸的能力。本研究筛选得到的菌株JK-4和JK-28具备较好的拮抗性能及植物促生潜力,为半夏绿色种植以及开发高效的生防菌剂提供了基础。

【Abstract】 To solve the disadvantages and economic losses caused by the bacterial soft rot disease in the planting of Pinellia ternata,we isolated two strains, namely JK-4 and JK-28,with strong antagonistic properties to Pectobacterium carotovorum subsp.carotovorum from the healthy rhizosphere soil collected from the planting cooperation of Pinellia ternata in Zhongxiang city, Hubei province.According to their morphological characteristics, physiological and biochemical characteristics and combined with their 16S rRNA sequence analysis, strain JK-4 and JK-28 were identified as Bacillus subtilis,and named as B.subtilis JK-4(JK-4) and B. subtilis JK-28(JK-28),respectively.Both JK-4 and JK-28 have strong abilities to secrete lipopeptide antibiotic surfactin.They produced 1.28 g/L and 1.12 g/L surfactin, respectively, when cultured in Landy medium at 30℃ and 200 r/min for 48 h.Additionally, they also could produce NH3,siderophores, ACC deaminase, and indole-3-acetic acid(IAA).In conclusion, the two strains isolated in this research showed both antagonistic properties to soft rot disease of Pinellia ternata and plant growth promoting potential, which provided the fundation for the development of effective biocontrol bacteria for Pinellia ternata planting.
