紫苏籽抗氧化肽对高脂饮食小鼠脂代谢及氧化应激的影响

Effects of Perilla Frutescens Seed Antioxidant Peptide on Lipid Metabolism and Oxidative Stress in Mice Fed High-Fat Diet

【摘要】 文章旨在探究紫苏籽抗氧化肽对高脂饮食小鼠脂代谢及氧化应激的影响。将6周龄雄性C57BL/6小鼠随机分为对照组、高脂组、高脂饮食+低剂量紫苏籽抗氧化肽、高脂饮食+高剂量紫苏籽抗氧化肽共4组,试验10周后,测定小鼠体质量、血清生化指标、肝脏甘油三酯(Triglyceride,TG)、总胆固醇(Total cholesterol, TC),并分析脂代谢及肠道紧密连接蛋白的关键基因等指标。紫苏籽抗氧化肽显著降低高脂小鼠体质量及肝脏指数,显著降低血清中谷草转氨酶(Aspartate transaminase, AST)和谷丙转氨酶(Alanine aminotransferase, ALT)、TG、TC、低密度脂蛋白(Low density lipoprotein, LDL-C)、D-乳酸(D-lactic acid, D-LA)、二胺氧化酶(Diamine oxidase, DAO)和血糖水平,显著提高了小鼠血清中高密度脂蛋白(High-density lipoprotein, HDL-C)水平,减少肝脏和肠道中活性氧(Reactive oxygen species, ROS)水平,提高了肝脏中超氧化物歧化酶(Superoxide dismutase, SOD)、过氧化物酶(Catalase, CAT)和谷胱甘肽过氧化物酶(Glutathione peroxidase, GSHPx)活性,降低肝脏中丙二醛(Malondialdehyde, MDA)水平。此外,紫苏籽抗氧化肽提高高脂饮食小鼠肠道中紧密连接蛋白1(Zonula occludens-1, ZO-1)和闭合蛋白(Occludin)mRNA的表达,降低肿瘤坏死因子α(Tumor necrosis factor alpha, TNF-α)m RNA的表达,且还提高了脂肪酸生成相关基因脂肪酸合成酶(Fatty acid synthetase, FAS)的表达,促进脂肪酸分解相关基因肉碱棕榈酰转移酶-1(Carnitine palmitoyl transferase-I, CPT-1)、脂蛋白脂酶(Lipoprotein lipase, LPL)和细胞色素P450家族成员7A1(Cytochrome P450 7A1, CYP7A1)的表达。紫苏籽抗氧化肽能显著缓解高脂饮食小鼠肥胖、减少脂质沉积和脂代谢异常,降低肠道氧化应激和维持肠道屏障完整性,并提高小鼠肝脏抗氧化能力,预防高脂饮食小鼠脂肪肝的发生。

【Abstract】 This artide investigated the effects of Perilla frutescens seed antioxidant peptides on lipid metabolism and oxidative stress in mice fed high-fat diet. Six week old male C57BL/6 mice were randomly divided into four groups: control group, high-fat group, high-fat diet+low dose Perilla frutescens seeds antioxidant peptide groups, and high-fat diet+high dose Perilla frutescens seeds antioxidant peptide groups. After 10 weeks of testing, body weight, serum biochemical indicators, liver triglyceride(TG), total cholesterol(TC), and key genes for lipid metabolism and intestinal tight junction proteins were measured in mice. Perilla frutescens seeds antioxidant peptides significantly reduced body weight and liver index in hyperlipidemic mice, significantly reduced serum aspartate transaminase(AST), alanine aminotransferase(ALT), TG, TC, low density lipoprotein(LDL-C), D-lactic acid(D-LA), diamine oxidase(DAO), and blood sugar level, significantly increased serum high-density lipoprotein(HDL-C) levels in mice, reduced the level of reactive oxygen species(ROS) in the liver and intestines; It increased the activities of superoxide dismutase(SOD), peroxidase(CAT), and glutathione peroxidase(GSH-Px) in the liver, and decreased the level of malondialdehyde(MDA) in the liver. In addition, Perilla frutescens seeds antioxidant peptides increased the expression of zonula occludens-1(ZO-1) and occlusin mRNA in the intestine of mice fed high-fat diet, and reduce tumor necrosis factor α(TNF-α) mRNA expression,and also increased the expression of fatty acid synthase(FAS), a fatty acid production related gene,and promoted the expression of carnitine palmitoyl transferase-1(CPT-1), lipoprotein lipase(LPL),and cytochrome P450 family member 7A1(CYP7A1). Perilla frutescens seed antioxidant peptides can significantly alleviate obesity, reduce lipid deposition and abnormal lipid metabolism in mice on high-fat diet, reduce intestinal oxidative stress, maintain intestinal barrier integrity, and improve the antioxidant capacity of the mice liver, preventing the occurrence of fatty liver in mice on a high fat diet.
