覆盆子中苷类物质的提取及抗炎活性研究
Extraction and Anti-Inflammatory Activity of Glycosides from Raspberry
【摘要】 以覆盆子作为原材料,在单因素实验的基础上,采用超声波辅助提取覆盆子中苷类物质,Box-Behnken响应面法优化提取工艺,应用电喷雾质谱(HPLC-ESI-MS)对覆盆子中苷类物质进行分析及评价覆盆子中苷类物质的抗炎作用。提取条件为料液比1:27 g/mL、乙醇体积分数57%、超声时间28 min、超声功率400 W,此时覆盆子苷类物质提取量为196.8 mg/100 g;MS/MS结果显示覆盆子中苷类物质主要有金丝桃苷、鞣花酸苷、槲皮素-3-O-芸香糖苷、槲皮素-3-O-葡萄糖醛酸苷、山奈酚-3-O-芸香糖苷、紫云英苷、椴树苷,其中鞣花酸苷含量最高;覆盆子苷类物质能够抑制由脂多糖(LPS)诱导的小鼠巨噬细胞RAW264.7与小鼠胚胎成纤维细胞NIH3T3的NO、PGE2分泌,验证覆盆子苷类物质具有很好的抗炎作用。
【Abstract】 Rraspberry was used as raw material. On the basis of single factor experiment, ultrasonic assisted extraction was used to extract glycosides from raspberry. Box-Behnken response surface methodology was adopted to optimize the extraction process. HPLC-ESI-MS was used to analyze the glycosides and evaluate the anti-inflammatory effect of the glycosides in raspberry. The extraction conditions were as follows: material liquid ratio 1:27 g/mL, ethanol volume fraction 57%, ultrasonic time 28 min, ultrasonic power 400 W. At this time, the extraction amount of raspberry glycosides was 196.8mg/100 g. MS/MS results indicated that the glycosides in raspberry mainly included hypericin, ellagic acid glycoside, quercetin-3-O-rutin, quercetin-3-O-glucuronic acid glycoside, kaempferol-3-O-rutin,shikonin and tillroside, among which ellagic acid glycoside was the highest. Raspberry glycosides can inhibit the secretion of NO and PGE2 in mouse macrophage RAW264.7 and mouse embryonic fibroblast NIH3T3 induced by lipopolysaccharide(LPS), which proves that raspberry glycosides have a good antiinflammatory effect.
