不消化性葡聚糖体外酵解液对RAW 264.7巨噬细胞的免疫调节作用

【摘要】 为探究不消化性葡聚糖(indigestible glucans,IGs)体外肠道菌酵解液的免疫调节活性,采用厌氧发酵模型利用健康人体粪便菌群分别对6种IGs(大麦β-葡聚糖、海带多糖、酵母β-葡聚糖、茯苓多糖、抗性淀粉和聚葡萄糖)进行24 h体外酵解,除菌过滤获得IGs酵解液,对pH值和短链脂肪酸含量进行测定。并以RAW 264.7巨噬细胞为研究对象,分为正常组、阳性组和IGs酵解液组,采用CCK-8试剂盒测定细胞活力,一氧化氮(nitric oxide,NO)检测试剂盒测定细胞NO释放量,流式细胞仪检测细胞活性氧(reactive oxygen species,ROS)含量,酶联免疫吸附测定法测定细胞因子分泌量。结果表明,相比于正常组,6种IGs的24 h酵解液均显著提高了RAW 264.7巨噬细胞活力、NO释放量、ROS产生量和小鼠肿瘤坏死因子α分泌量。各IGs 24 h肠道菌酵解液对RAW 264.7巨噬细胞均具有免疫调节的作用。

【Abstract】 To investigate the immunomodulatory activity of intestinal bacterial in vitro fermentation broths of indigestible glucans(IGs),six IGs(barley β-glucan,laminarin,yeast β-glucan,pachyman,resistant starch,and litesse) were fermented in vitro for 24 h by anaerobic fermentation model using healthy human fecal microbiota,and IG fermentation broths were obtained by sterile filtration. pH and short-chain fatty acid levels were measured. RAW 264.7 macrophages were divided into normal,positive,and IG fermentation broth groups.Cell viability was determined with a CCK-8 kit,and levels of nitric oxide(NO),reactive oxygen species(ROS),and cytokines were analyzed using NO detection kit,flow cytometry,and enzyme-linked immunosorbent assay,respectively. The results suggested that compared with the normal group,all 24 h fermentation broths of six IGs were able to enhance the viability of RAW 264.7 macrophages,production of NO and ROS,and TNF-α release.The intestinal bacterial fermentation broth of each IG had the immunomodulatory ability on RAW 264.7macrophages.
