4种消除高粱直链淀粉测定中支链淀粉干扰的方法比较

【摘要】 基于比色法原理,比较4种不同测定方法对支链淀粉干扰的消除作用。采用分光光度计全扫描模式,建立测定方法,设计验证试验比较不同测定方法对支链淀粉干扰的消除作用,并选择适合方法对7个高粱样品进行测定。根据全扫描结果,确定测定波长和标准曲线,结果均满足测定要求。通过验证试验发现, 740 nm混标单波长法(方法二)和混标双波长法(方法三)可有效地消除支链淀粉干扰。对高粱样品测定发现,除糯高粱样品,方法二和方法三测定结果差异不显著,在设定混合标样总淀粉含量为70%的体系中,混标双波长法不适合测定糯性高粱样品。740 nm混标单波长法可有效消除支链淀粉干扰,计算简单,适合高粱直链淀粉测定。

【Abstract】 Based on the principle of colorimetry, compare the elimination effects of four different determination methods on amylopectin interference. The determination method was established by using the full scan mode of spectrophotometer, a validation test was designed to compare the elimination effect of different determination methods on amylopectin interference,and a suitable method was selected for the determination of 7 sorghum samples. According to the full scan results, the determination wavelength and standard curve were determined to meet the determination requirements; The validation test showed that 740 nm mixed standard single wavelength method(method 2) and mixed standard dual wavelength method(method 3) could effectively eliminate amylopectin interference; The determination of sorghum samples showed that there was no significant difference between the determination results of method 2 and method 3 except waxy sorghum samples. The 740 nm mixed standard dual wavelength method could effectively eliminate the interference of amylopectin, and was simple calculation. It was suitable for the determination of sorghum amylose. The mixed standard dual wavelength method was not suitable for the determination of waxy sorghum samples in the system where the total starch content of the mixed standard sample was set at 70%.
