猪肝中瘦肉精的TLC-SERS检测方法研究
Study of Determination Method for TLC-SERS of Lean Meat Powder in Pig Liver
【摘要】 目的:建立薄层色谱与表面增强拉曼光谱联用技术检测猪肝中3种瘦肉精的新方法。方法:先将盐酸克仑特罗、盐酸班布特罗和富马酸福莫特罗等3种对照品及处理后的猪肝样品分别用甲醇配制对照品溶液和供试品溶液,再建立薄层色谱分离系统,在254 nm下检视定位后在相应组分斑点处滴加表面增强剂银溶胶后检测其拉曼光谱,并对其进行灵敏度验证,最后对市售猪肝进行检测。结果:薄层色谱与表面增强拉曼光谱联用技术可显著增强拉曼光谱检测信号。盐酸克仑特罗、盐酸班布特罗和富马酸福莫特罗的检测限分别为1μg、0.5μg和0.5μg。结论:本法灵敏度高,为苯乙胺类拟肾上腺素药物残留物的鉴别技术提供了一种快速、高效的新方法。
【Abstract】 Objective: To establish a new method for the detection of 3 kinds of lean meat powder in pig liver by thin-layer chromatography(TLC) and surface-enhanced Raman spectroscopy(SERS). Method: Firstly, clenbuterol hydrochloride, bambuterol hydrochloride, formoterol fumarate and the treated pig liver samples were prepared with methanol as the reference solution and the test solution, and then a TLC separation system was established. After examining and positioning at 254 nm, the surface enhancer silver sol was added to the spots of the corresponding components to detect their Raman spectra and verify their sensitivity. Finally, the commercial pig liver was tested.Result: TLC combined with SERS could significantly enhance the detection signal of Raman spectrum. The detection limits of clenbuterol hydrochloride, bambuterol hydrochloride and formoterol fumarate were 1 μg, 0.5 μg and 0.5 μg, respectively. Conclusion: This method has high sensitivity, and provides a rapid and efficient new method for the identification of phenethylamines and adrenergic drug residues.
