窖泥产氨菌的分离筛选与发酵培养基的优化

【摘要】 为获取可提升窖泥pH的高产氨态氮菌株,本文以优质老窖泥为菌源,采用多次富集培养的方式,从中筛选到一株高产氨态氮的菌株J19。对菌株进行形态学观察和分子生物学鉴定;并采用正交设计对氨态氮发酵培养基组分进行优化。结果表明,菌株J19为陆地寡养单胞菌(Stenotrophomonas terrae);发酵培养最佳组分为蛋白胨5.0 g/L,K2HPO4·3H2O 0.20 g/L,MgSO4·7H2O 0.50 g/L,FeSO4·7H2O 0.010 g/L,氨态氮产量为232.176 mg/L。

【Abstract】 In order to obtain a high ammonium nitrogen-producing strain that can raise the pH of cellar mud,this paper used high quality old cellar mud as the source of bacteria,and used multiple enrichment culture,from which a high ammonium nitrogen-producing strain J19 was selected.Morphological observation and molecular biology identification of the strains.An orthogonal design was also used to optimize the ammonium nitrogen fermentation medium components.The results showed that strain J19 was Stenotrophomonas terrae,The best medium was peptone 5.0 g/L,K2HPO4·3H2O 0.20 g/L,MgSO4·7H2O 0.50 g/L,FeSO4·7H2O 0.010 g/L,and the ammonia nitrogen content was 232.176 mg/L.
