一株野生树舌灵芝菌株鉴定及其液态发酵特性

【摘要】 为进一步丰富灵芝真菌资源,对从野外桃树上采集的灵芝子实体,采用形态学观察和ITS序列比对进行菌种鉴定,并对其摇瓶发酵液的菌丝体生物量和胞内外代谢物特性进行分析。结果表明:该灵芝菌株在分类上属于多孔菌科灵芝属,学名为树舌灵芝,序列上传Gen Bank,登录号为OK037622。液态发酵研究表明,其菌丝体生物量为15.16 g∕L,底物葡萄糖利用率为77.63%,胞内外多糖含量和质量浓度分别为16.04 mg∕g和1.23 g∕L,胞内代谢物主要有糖醇类(45.07%)、糖类(40.05%)、脂肪酸类(13.33%),胞外代谢物主要有氨基酸类(40.02%)、糖类(15.74%)、糖酸类(12.09%)、糖醇类(9.85%)、脂肪酸类(6.28%)、有机酸类(5.82%)、酯类(4.34%)、醇类(2.83%)、核苷类(2.79%)。该研究可为下一步开展灵芝研究提供菌种准备,并为后续的开发应用提供参考。

【Abstract】 In order to further enrich the fungal resources ofGanoderma,the fruiting bodies ofGanoderma collected from wild peach trees were identified by morphological observation and ITS sequence alignment. The mycelial biomass and characteristics of intracellular and extracellular metabolites in the shake flask fermentation broth were also analyzed. The results showed that theGanoderma lucidumstrain belongs to the Polyporaceae,Ganoderma,with the scientific name ofG. applanatum. The sequence was uploaded to GenBank with the accession number OK037622. Liquid fermentation studies showed that its mycelial biomass was 15. 16 g∕L,the substrate glucose utilization rate was 77. 63%,the content and concentration of intracellular and extracellular polysaccharides were 16. 04 mg∕g and 1. 23 g∕L,respectively. The intracellular metabolites mainly included sugar alcohols(45. 07%),saccharides(40. 05%),and fatty acids(13. 33%). The extracellular metabolites mainly included amino acids(40. 02%),saccharides(15. 74%),sugar acids(12. 09%),sugar alcohols(9. 85%),fatty acids(6. 28%),organic acids(5. 82%),esters(4. 34%),alcohols(2. 83%),and nucleosides(2. 79%). This study could provide strain preparation for the next step ofGanodermaresearch, and provide reference for subsequent development and application.
