基于PI3K/Akt/mTOR信号通路的苦参碱诱导人乳腺癌MCF-7细胞自噬及其机制的研究

【摘要】 目的 研究苦参碱对人乳腺癌MCF-7细胞自噬及其相关信号通路的影响,为苦参碱的抗肿瘤作用提供更多依据。方法 采用自噬双标腺病毒(mRFP-GFP-LC3)法、透射电镜法(TEM)检测给药后细胞自噬水平的变化;Western blot检测苦参碱对自噬相关蛋白LC3、Beclin-1,信号通路蛋白PI3K、p-PI3K、Akt、p-Akt、mTOR、p-mTOR表达水平的影响。结果 随着苦参碱浓度的上升,细胞中的自噬水平也在逐渐升高,自噬相关蛋白LC3-II/LC3-I、Beclin-1的表达量呈上升趋势,通路相关蛋白PI3K、Akt、mTOR无明显变化,p-PI3K、p-Akt、p-mTOR的表达量均呈现下降趋势。结论 苦参碱可剂量依赖性升高MCF-7细胞的自噬水平,机制可能与PI3K/Akt/mTOR介导的信号通路有关。
【Abstract】 Objective To study the effects of matrine on autophagy and related signaling pathways of human breast cancer MCF-7 cells, and to provide more evidence for the antitumor effect of matrine. Methods Autophagy double labeled adenovirus(mRFP-GFP-LC3) and transmission electron microscopy(TEM) were used to detect the changes of autophagy level. Western blot was used to detect the effect of matrine on the expression of autophagy related protein LC3, Beclin-1, and signal pathway proteins including PI3K, p-PI3K, Akt, p-Akt, mTOR and p-mTOR. Results With the increase of matrine level, the autophagy level in cells increased gradually. The expression of autophagy related proteins LC3-II/LC3-I and Beclin-1 showed an upward trend. The expression of pathway related proteins PI3K, Akt and mTOR did not change significantly. The expression of p-PI3K, p-AKT and p-mTOR showed a downward trend.Conclusion Matrine can increase the autophagy level of MCF-7 cells in a dose-dependent manner. The mechanism may be related to the PI3K/Akt/mTOR mediated signal pathway.
