基于网络药理学与分子对接技术探讨天麻治疗高血压的作用机制
Research of Potential Mechanisms of Gastrodia elata Blume in the Treatment of Hypertension Based on Network Pharmacology and Molecular Docking
【摘要】 目的:本研究采用网络药理学及分子对接方法探讨天麻治疗高血压作用的分子机制。方法:天麻的主要活性成分来自TCMID和Pubchem数据库。利用OMIM,Disgenet, Genecards数据库建立了高血压靶点数据库。利用Cytoscape软件(3.7.2版)和R3.6.1软件建立“药物-成分-靶点-疾病”网络图。利用STRING数据库构建了蛋白-蛋白相互作用(PPI)网络,并分析了蛋白相互作用关系。对核心靶点进行GO生物功能分析和KEGG富集分析。采用分子对接验证实验进行了验证。结果:天麻中的32种活性成分可以调控高血压的118个靶点。前5位关键活性成分为4-(4’-hydroxybenzyloxy)benzyl methyl ether, 4-ethoxymethylphenyl-4’-hydroxybenzylether, dauricine, 20-Hexadecanoylingenol, suchilactone,关键靶点为AKT1、ALB、NOS3、VEGFA、SRC。网络分析结果表明,这些靶点主要参与调节循环系统中的血管过程、调节管径、调节血管大小、调节血管直径、调节管径、调节血压等生物过程。参与高血压治疗的通路包括ERBB信号通路、Jak-STAT信号通路、PI3K/AKT信号通路、mTOR信号通路。分子对接验证实验结果表明,前5位化合物与AKT1、ALB、NOS3、VEGFA、SRC具有较好的结合活性。结论:天麻治疗高血压具有多成分作用于多靶点的特点,其作用机制涉及调节血浆胶体渗透压、细胞增殖和血管生成、炎症反应。本研究为进一步开展天麻治疗高血压的作用机制研究提供了新思路。
【Abstract】 Objective: This study used a network pharmacology approach to elucidate the molecular mechanism governing the effect of Rhizome of Gastrodia elata Blume(RGE) on hypertension.Methods: The main active ingredients of RGE were obtained from Traditional Chinese Medicines Integrated Database(TCMID) and Pubchem Database.The GeneCard, OMIM and Disgenet database were used to establish a database of hypertension targets.The interactive active network map of “ingredient-target” was constructed with Cytoscape software(Version 3.7.2) and software R3.6.1.The protein-protein interaction network was constructed with the STRING database and the related protein interaction relationship was analysed.GO biological function analysis and KEGG enrichment analysis for core targets were performed.Molecular docking validation experiments as further and precise research were adopted.Results: Thirty-two active ingredients of RGE were obtained and 118 potential common targets for “RGE-hypertension” were identified.Potential targets mainly involved in AKT1,ALB,NOS3,VEGFA,SRC.Network analysis results indicate that these targets are primarily involved in regulating biological processes, such as vascular process in circulatory system, regulation of tube diameter, regulation of blood vessel size, regulation of blood vessel diameter, regulation of tube size, regation of blood pressure.Pathways involved in the treatment of hypertension include ERBB signaling pathway, Jak-STAT signaling pathway, PI3K/AKT signalling pathway, mTOR signalling pathway.Molecular docking validation experiments results showed that 5 compounds including 4-(4’-hydroxybenzyloxy)benzyl methyl ether, 4-ethoxymethylphenyl-4’-hydroxybenzylether, dauricine, 20-Hexadecanoylingenol and suchilactone had better binding activity with AKT1,ALB,NOS3,VEGFA,SRC through molecular docking.Conclusion: RGE has the characteristics of being multi-component and multi-target.Possible mechanisms of action for RGE include regulating of blood plasma colloid osmotic pressure, cell proliferation and angiogenesis, inflammatory response.This study provides a new idea for further research on the mechanism of Gastrodia elata Blume in the treatment of hypertension.
