金钱鱼毒腺基因NeoVTX的基因序列和表达
Gene Sequence and Expression of NeoVTX from Venom of Scatophagus argus
【摘要】 为了解金钱鱼毒液成分,从其基因组中分离了NeoVTX-α和NeoVTX-β基因序列,进行生物信息学分析,并研究它们在毒腺及其他组织的表达特征。结果表明,金钱鱼NeoVTX-α和NeoVTX-β的开放阅读框ORF全长分别为1 581 bp和2 106 bp,分别编码526和701个氨基酸。同源性和系统进化分析发现,金钱鱼NeoVTX-α与鲉形目的荫平鲉、尖头拟鲉、玫瑰毒鲉、粗毒鲉等鱼类同属一支且同源性较高(67.3%~73.5%)。金钱鱼NeoVTX(α和β亚基)三维结构与粗毒鲉的SNTX(α和β亚基)毒素的三维结构比较相似。qPCR和转录组数据表明,金钱鱼NeoVTX-α和NeoVTX-β在毒腺组织中高表达,其他组织低表达或不表达。研究表明,NeoVTX可能是金钱鱼中一种与鲉形目毒素相类似的毒素。
【Abstract】 To understand the role of NeoVTX in venom gland of Scatophagus argus,the NeoVTX-α and NeoVTX-βgene sequences were isolated from the genome for bioinformatics analysis and their expression characteristics in the venom gland and other tissues were investigated.The results showed that the full length of the open reading frame(ORF) of NeoVTX-α and NeoVTX-β is 1 581 bp and 2 106 bp,encoding 524 and 701 amino acids,respectively. Homology and phylogenetic analysis revealed that NeoVTX-α belongs to the same family and has high homology(67.3%~73.5%) with Sebastes umbrosus,Scorpaenopsis oxycephala,Synanceia verrucosa,and Synanceia horrida. The 3D structure of Scatophagus argus NeoVTX(α and β subunits) is similar to that of SNTX toxin(α and β subunits) of Synanceia horrida. The qPCR and transcriptome data indicated that NeoVTX-α and NeoVTX-β was highly expressed in toxic glands of Scatophagus argus,while other tissues showed low or no expression. The data suggest that NeoVTX in the Scatophagus argus may be a toxin similar to that in Scorpaeniformes.
