Piezo1激活对尿路上皮细胞系Notch配体表达的影响
The effects of Piezo1 activation on the expression of Notch ligands in urothelial cell line
【摘要】 目的 探究膀胱出口梗阻之后,机械敏感性离子通道Piezo1激活对尿路上皮细胞系Notch配体表达的影响。方法 分别使用高静水压或Piezo1通道激动剂Yoda1,对SV-HUC-1尿路上皮细胞系进行处理;使用Western blot实验检测Notch配体蛋白的表达水平。结果 在使用高静水压或Yoda1处理之后,SV-HUC-1细胞系Notch配体蛋白的表达水平均有不同程度的上升;Piezo1拮抗剂Dooku1能部分抑制高静水压激活Piezo1通道导致的Notch配体蛋白表达增加。结论 激活Piezo1通道可导致SV-HUC-1细胞系中Notch配体蛋白表达上升,这可能参与了膀胱出口梗阻后膀胱结构重塑以及膀胱功能受损的进程。
【Abstract】 Objective This study aimed to investigate the effects of activation of the mechanosensitive ion channel Piezo1 on the expression of Notch ligands in urothelial cell line following bladder outlet obstruction. Methods SV-HUC-1 was treated with high hydrostatic pressure or the Piezo1 channel agonist Yoda1. The expression levels of Notch ligands were detected by Western blot. Results After treatment with high hydrostatic pressure or Yoda1, the expression of Notch ligands in SV-HUC-1 increased. Furthermore, the Piezo1 antagonist Dooku1 inhibited the increase in the expression of Notch ligands induced by high hydrostatic pressure. Conclusions The activation of Piezo1 channel increases the expression of Notch ligands in SV-HUC-1,which may be involved in the process of bladder remodeling and bladder dysfunction due to bladder outlet obstruction.
