基于网络药理学的半枝莲治疗膀胱癌相关作用机制研究
Study on the related mechanism of Scutellariae Barbatae Herba against bladder cancer based on network pharmacology
【摘要】 目的 通过网络药理学方法探讨半枝莲治疗膀胱癌的作用机制,为临床治疗提供理论依据。方法 利用中药系统药理数据库和分析平台(TCMSP)数据库获得半枝莲化学成分及对应作用靶点基因,利用Gene Cards数据库获得膀胱癌相关靶点基因,利用Cytoscape软件构建半枝莲“成分-靶点-疾病”三者相互关系网络,采用STRING数据库对半枝莲作用于膀胱癌的潜在作用靶点进行蛋白相互作用网络分析并筛选关键靶点。利用在线工具Metascape对共同靶点进行基因本体功能富集分析和京都基因和基因组百科全书(KEGG)信号通路富集分析。结果 半枝莲含29个活性成分、228个靶基因,膀胱癌含4 645个靶基因,共同靶点190个,关键靶点可能为AKT1、TP53、JUN、TNF、EGFR、MAPK1等。KEGG富集分析主要涉及癌症途径、糖尿病并发症中的AGE-RAGE信号通路、PI3K-Akt信号通路等。结论 本研究借助网络药理学方法初步预测了半枝莲通过多靶点、多通路治疗膀胱癌,为临床治疗提供新靶点和新思路。
【Abstract】 Objective To study the related mechanism of Scutellariae Barbatae Herba against bladder cancer based on network pharmacology. Methods Chemical constituents and action target genes of Scutellariae Barbatae Herba were obtained from TCMSP database, while Gene Cards was retrieved for the target genes of bladder cancer. Cytoscape software was used to construct the interrelationship network of compound-target-disease. The protein interaction network of common target genes was analyzed using STRING database, and Cytoscape was used to screen the key genes. Gene Ontology(GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway analysis were conducted using Metascape online tool. Results The screening revealed that Scutellariae Barbatae Herba contained 29 chemical constituents and 228 target genes. Bladder cancer contained 4 645 target genes. One hundred and ninety potential targets were identified for the treatment of bladder cancer with Scutellariae Barbatae Herba. The key genes could be AKT1, TP53, JUN, TNF, EGFR, MAPK1, etc. The KEGG enrichment analysis mainly involved the cancer pathway, AGE-RAGE signaling pathway, PI3K-Akt signaling pathway and other pathways. Conclusions With the help of network pharmacology, this study preliminarily predicted that Scutellariae Barbatae Herba could treat bladder cancer through multi-target and multi-pathway.
