槲皮素及其衍生物抗植物病毒的初步研究
The Preliminary Study on Quercetin and Its Derivatives Aganist Plant Virus
【摘要】 采用半叶枯斑法和酶联免疫法测试槲皮素及其衍生物对烟草花叶病毒、黄瓜花叶病毒、马铃薯Y病毒和水稻条纹病毒的抑制活性,并采用RT-qPCR的方法研究槲皮素及其衍生物对Hsp mRNAs表达的影响。研究结果表明,槲皮素及其衍生物7,4′-Di-O-甲基槲皮素、3′-O-甲基-6-氢-槲皮素、3-O-甲基槲皮素、3,7,4′-Tri-O-甲基槲皮素和3,7,3′,4′-Tetra-O-甲基槲皮素在100μmol/L的浓度下对4种病毒具有较强的抑制活性。7,4′-Di-O-甲基槲皮素对4种病毒的抑制活性分别为68.46%、51.10%、62.18%和58.05%,3-O-甲基槲皮素对其抑制活性分别为52.47%、54.82%、48.55%和46.52%。42℃高温热激协同槲皮素处理能下调Hsp70和Hsp90 mRNA,并在125~1 000μmol/L的作用剂量范围内呈剂量依赖关系。槲皮素对Hsp90 mRNA的下调效果比Hsp70 mRNA显著。42℃热激处理本氏烟Hsp70mRNA相对表达倍数为3.59倍,槲皮素、7,4′-Di-O-甲基槲皮素、3-O-甲基槲皮素和7-O-甲基槲皮素与热激协同处理,Hsp70 mRNA表达值分别下调48.74%、66.30%、37.05%和22.84%,即7,4′-Di-O-甲基槲皮素对下调Hsp70 mRNA的效果最为明显。本研究结果为今后筛选高活性的抗病毒物质提供指导。
【Abstract】 The activities of quercetin and its derivatives against tobacco mosaic virus (TMV),cucumber mosaic virus(CMV),potato Y virus (PVY) and rice stripe virus (RSV) were tested by the half-leaf method and enzyme-linked immunoassay.The expression abundance of m RNA of heat shock protein (Hsp)70 and Hsp90 in Nicotiana benthamiana leaves induced by quercetin and its derivatives were studied by RT-qPCR.The results showed that quercetin and its derivatives7,4’-Di-O-methyl quercetin,3’-O-methyl-6-hydrogen quercetin,3-O-methyl quercetin,3,7,4’-Tri-O-methyl quercetin and3,7,3’,4’-Tetra-O-methyl quercetin possessed stronger inhibitory effect against four viruses at the dosage of 100μmol/L.The inhibitory effects of 7,4’-Di-O-methylquercetin against four viruses were 68.46%,51.10%,62.18%and 58.05%,and the inhibitory effects of 3-O-methylquercetin were 52.47%,54.82%,48.55%and 46.52%,respectively.Quercetin coupled with heat shock treatment at 42℃could significantly down-regulate Hsp70 and Hsp90 m RNAs in a dose-dependent manner from 125μmol/L to 1 000μmol/L.The down-regulation effect of quercetin on Hsp90 m RNA was more significant than that of Hsp70 m RNA.The relative expression values of m RNA Hsp70 in tobacco leaves under heat shock was 3.59.The down-regulation fold of quercetin,7,4’-Di-O-methylquercetin,3-O-methylquercetin and 7-O-methylquercetin on Hsp70m RNA expression were 48.74%,66.30%,37.05%,and 22.84%,respectively.7,4’-Di-O-methyl quercetin could significantly down-regulate Hsp70 m RNA.The results provide guidance for screening highly active antiviral substances in the future.
