【摘要】 目的 探讨lncRNA DIO3OS在骨肉瘤中的表达及其在调控骨肉瘤细胞生物学行为中的作用。方法 收集骨肉瘤标本及配对癌旁正常组织标本6例，qRT-PCR检测骨肉瘤组织中DIO3OS表达。MG-63细胞转染si-DIO3OS或pcDNA3.1-DIO3OS,敲降或过表达DIO3OS后，通过CCK-8实验评估细胞增殖能力，划痕实验和Transwell实验检测骨肉瘤细胞MG-63迁移与侵袭能力。基于TARGET数据库，根据患者骨肉瘤组织中DIO3OS表达水平分为高表达组与低表达组，Kaplan Meier生存分析评估DIO3OS表达水平与患者预后的关系。结果 qRT-PCR结果显示，骨肉瘤组织及细胞中DIO3OS表达下调。敲降DIO3OS表达时，CCK-8实验结果表明骨肉瘤细胞MG-63增殖能力增加，划痕实验和Transwell实验结果显示MG-63细胞迁移与侵袭能力增加(P<0.05)。过表达DIO3OS后，CCK-8、划痕实验和Transwell实验结果显示，MG-63细胞的增殖、迁移与侵袭能力减弱(P<0.05)。Kaplan-Meier生存分析结果显示，DIO3OS高表达组患者生存率明显高于低表达组(P<0.05)。结论 lncRNA DIO3OS在骨肉瘤组织及细胞中表达下调，并且可以抑制骨肉瘤细胞的增殖、迁移与侵袭能力，DIO3OS高表达的骨肉瘤患者预后较好。更多还原

【Abstract】 Objective To investigate the expression of LncRNA DIO3OS in osteosarcoma and the role of it in regulating the biological behavior of osteosarcoma cells.Methods 6 cases of osteosarcoma tissues were collected and DIO3OS expression was detected by qRT-PCR. The ability of proliferation, migration and invasion of MG-63 cells were evaluated by CCK-8,scratch assay and transuell assay after transfecting si-DIO3OS or pcDNA3.1-DIO3OS. The overall survival curve was drawn to evaluate the relationship between DIO3OS expression level and the prognosis of patients based on the Kaplan-Meier survival analysis of the TARGET database.Results The expression of DIO3OS in osteosarcoma tissues and cells was dounregufed compared to the normal control tissue and cell. The ability of proliferation, migration and invasion of MG-63 cells were increased after the knockdown of DIO3OS as determined by CCK-8,scratch assay and transwell assay(P<0.05). The ability of proliferation, migration and invasion of MG-63 cells were decreased after DIO3OS overexpression(P<0.05). The Kaplan-Meier analysis showed that the survival rate of patients was significantly higher than that of patients with low expression(P<0.05).Conclusions LncRNA DIO3OS was dounregufed in osteosarcoma tissues and cells. LncRNA DIO3O Sinhibited the proliferation, migration and invasion of osteosarcoma cells. Osteosarcoma patients with high expression of DIO3O S had a better prognosis.更多还原
