基于正交试验的地菍扦插繁殖研究
Study on cutting propagation of Melastoma dodecandrum based on orthogonal experiment method
【摘要】 以地菍(Melastoma dodecandrum)当年生半木质化枝条为插穗,蛭石为基质,采用正交试验和主成分分析,研究生根剂种类、生根剂浓度及处理时间的不同水平和处理组合条件下对地菍扦插生根效果的影响,筛选出适合地菍扦插繁殖的方案,旨在高效培育地菍幼苗,为地菍的扦插繁殖提供技术和理论的支撑。结果表明:地菍插穗以皮层生根和结上生根为主,不同处理组合的扦插生根率为40.00%~86.67%,平均根数为13.17~36.22条/株,平均根长为1.15~2.43 cm,最长根长为1.37~5.22 cm,生根效果指数为6.58~21.09,各处理间对地菍插穗生根影响均较大,且不同处理之间差异显著(P<0.05)。极差分析发现:生根剂浓度是影响地菍平均根长和最长根长的主要因素;生根剂种类是影响地菍平均生根数、生根效果指数的主要因素;影响地菍生根率的主导因子为生根剂处理时间。主成分分析联合生根效果指数指标极差分析结果得到,500 mg/L NAA处理0.5 min和100 mg/L IBA处理30 min是较适合地菍插穗生根的组合。
【Abstract】 Using semi-lignified branches of Melastoma dodecandrum as cuttings and vermiculite as matrix, orthogonal test and principal component analysis were used to study the effects of different levels and combinations of rooting agents, rooting agent concentration and treatment time on the rooting effect of Melastoma dodecandrum cuttings. The suitable cutting propagation scheme of M. dodecandrum was screened to cultivate M. dodecandrum seedlings efficiently and provide technical and theoretical support for cutting propagation of M. dodecandrum.The results showed that cortex rooting and supranodal rooting were the main rooting types of M. dodecandrum cuttings. The rooting rate of different treatment combinations 40.00%~86.67%, the average number of roots was 13.17~36.22 per plant, the average root length was 1.15~2.43 cm, the longest root length was 1.37~5.22 cm, and the rooting effect index was 6.58~21.09. All treatments had great effects on rooting of M. dodecandrum cuttings. There was significant difference among different treatments(P<0.05). Range analysis found that rooting agent concentration was the main factor affecting the average root length and longest root length of M. dodecandrum. The types of rooting agents were the main factors affecting the average rooting number and rooting effect index of M. dodecandrum. The main factor affecting the rooting rate of M. dodecandrum was the treatment time of rooting agent. The results of principal component analysis combined with the range analysis of rooting index showed that 500 mg/L NAA for 0.5 min and 100 mg/L IBA for 30 min were more suitable for rooting of M. dodecandrum cuttings.
