铁代谢相关基因FLVCR1对肝细胞癌的预后影响及与免疫细胞浸润的相关性研究

【摘要】 目的 基于多个数据库探索猫白血病病毒C亚群受体1(feline leukemia virus subgroup C receptor 1,FLVCR1)在肝细胞癌(Hepatocellular carcinoma, HCC)中的表达、预后及与免疫细胞浸润的相关性。方法 利用UALCAN、GEPIA2、仙桃学术平台分析FLVCR1在HCC患者中的表达情况;利用GEPIA、GSCA分析FLVCR1与临床分期的关系;Linkedomics寻找与FLVCR1相关的基因,进一步进行GO、KEGG富集分析;利用TIMER数据库分析FLVCR1与免疫细胞浸润的相关性;Kaplan Meier plotter探索免疫细胞浸润高低对预后的影响;在GSCA分析FLVCR1与药物敏感性的相关性。结果 FLVCR1在HCC患者中高表达;FLVCR1表达与临床分期呈正相关;与FLVCR1正相关的基因主要富集在有机酸分解代谢等过程,与FLVCR1负相关的基因主要富集在染色体分离等过程;FLVCR1表达水平与四种免疫细胞包括CD4~+记忆T细胞、CD8~+T细胞、髓源性抑制细胞(myeloid-derived suppressor cells, MDSCs)、调节性T细胞(regulatory cells, Tregs)的浸润水平呈正相关且P<0.05;Kaplan Meier plotter数据库结果说明四种免疫细胞浸润与HCC患者不良预后相关;FLVCR1的表达与Dasatinib药物敏感性正相关,与Navitoclax负相关。结论 FLVCR1对HCC患者的不良预后与免疫细胞浸润密切相关。

【Abstract】 Objective To explore the expression and prognosis of FLVCR1 in hepatocellular carcinoma(HCC) and its correlation with immune cell infiltration based on multiple databases.Methods The UALCAN, GEPIA2 and Xiantao academic platform were used to analyze the expression of FLVCR1 in HCC.GEPIA and GSCA were used to analyze the relationship between FLVCR1 and clinical stage.The genes related to FLVCR1 were searched by Linkedomics, and GO and KEGG enrichment analysis were further conducted.TIMER database was used to analyze the correlation between FLVCR1 and immune cell infiltration.Kaplan Meier Plotter was used to explore the effect of immune cell infiltration on prognosis.The correlation between FLVCR1 and drug sensitivity was analyzed in GSCA.Results FLVCR1 was highly expressed in HCC patients.The FLVCR1 expression was positively correlated with clinical stage.Genes positively correlated with FLVCR1 were mainly enriched in organic acid catabolism and other processes.The genes negatively correlated with FLVCR1 were mainly enriched in the process of chromosome segregation.The expression level of FLVCR1 was positively correlated with the infiltration levels of four immune cells, including CD4~+ memory T cells, CD8~+ T cells, MDSCs and Tregs(P<0.05).Kaplan Meier Plotter database results showed that high infiltration of four types of immune cells was associated with poor prognosis in patients with HCC.FLVCR1 expression was positively correlated with Dasatinib drug sensitivity and negatively correlated with Navitoclax.Conclusion The poor prognosis of FLVCR1 in patients with HCC is closely related to immune cell infiltration.
