响应面法优化麦冬黄酮纯化工艺及其对运动疲劳的影响

【摘要】 以麦冬黄酮的吸附率和解吸率为考察指标,探讨HPD-300大孔树脂纯化麦冬黄酮的最佳工艺条件,并通过小鼠的运动疲劳模型,比较不同产物缓解运动疲劳的活性。结果表明:最佳纯化条件为上样液质量浓度0.4 mg/mL、上样液pH4.1、上样流速0.9 mL/min、上样体积50 mL、洗脱液体积100 mL(体积分数70%乙醇溶液)和洗脱流速1.9 mL/min,所得产物的总黄酮含量由18.2%±0.5%提高至69.4%±1.2%。与空白对照组和提取组相比,灌胃高剂量(0.4 g/kg·d)麦冬黄酮纯化物的小鼠游泳力竭时间显著延长(P<0.05),同时运动后血清中乳酸与尿素氮的含量显著下降(P<0.05)。

【Abstract】 Using the adsorption and desorption rates of flavonoids from Ophiopogon japonicus as indicators, the optimal process conditions for purifying flavonoids from Ophiopogon japonicus using HPD-300 macroporous resin were explored. The activity of different products in alleviating exercise fatigue was compared using a mouse exercise fatigue model. The results showed that the optimal purification conditions were as follows: sample solution mass concentration 0.4 mg/mL, sample solution pH 4.1, flow rate 0.9 mL/min, sample volume 50 mL, eluent volume 100 mL(volume fraction 70 % ethanol solution), and flow rate 1.9 mL/min. The total flavonoid content of the obtained product increased from 18.2 %±0.5 % to 69.4 %±1.2 %. Compared with blank control group and extraction group, the swimming exhaustion time of mice administrated with high dose(0.4 g/kg·d) of flavonoids purified from Ophiopogon japonicus was significantly extended(P<0.05), and the contents of lactic acid and urea nitrogen in serum were significantly decreased after exercise(P<0.05).
