赤斑羚皮肤成纤维细胞的分离和培养

【摘要】 研究旨在探索和建立稳定的赤斑羚皮肤成纤维细胞的体外分离、培养的技术方法,为后续赤斑羚的保护与饲养提供医学基础,并利用该细胞开展赤斑羚皮肤疾病的致病机制研究和相关药物研发。取赤斑羚(Naemorhedus baileyi)后腿内侧皮肤用Ⅰ型胶原酶进行分离培养,建立赤斑羚皮肤组织成纤维细胞系,并对其相关生物学特性进行鉴定。结果显示,试验所建立的赤斑羚成纤维细胞系大多数细胞呈细长梭状,部分细胞呈三角状。细胞多为放射状和漩涡状分布,排列整齐。培养过程中,细胞在贴壁1 d时开始有细胞向外伸展,贴壁10 d后细胞密度达到80%以上,可进行传代;细胞长势良好,生长曲线呈典型的S形;细胞复苏前后活率无显著差异,显示细胞生长状态好。研究表明,试验建立的赤斑羚成纤维细胞系为后续赤斑羚的相关研究奠定了基础。

【Abstract】 The purpose of the study was to explore and establish a technique for the isolation and culture of stable skin fibroblasts in vitro, so as to provide a medical basis for the subsequent protection and feeding of impala, and to use these cells to carry out research on the pathogenic mechanism of skin diseases of Impala and related drug development. The inner skin of the hind legs of Naemorhedus baileyi was isolated and cultured with collagenase type I to establish the skin tissue fibroblast cell line of Naemorhedus baileyi. The results showed that most of the cells were spindle-like, and some of the cells were triangular. The cells were mostly distributed radially and swirly, and arranged neatly. In the process of culture,the cells began to extend outward at 1 day of adhesion, and the cell density reached more than 80% after 10 days of adhesion, and the cells could be passed. The cell growth was good, and the growth curve was typical S-shape. There was no significant difference in cell viability before and after resuscitation, indicating good cell growth. The study indicates the fibroblast cell lines established by the method lay a foundation for the subsequent studies on the Naemorhedus baileyi.
