万年青炭疽病菌的识别与鉴定
Identification of Rohdea japonica Anthracnose Causal Agent
【摘要】 【目的】鉴定高邮市普遍发生的万年青(Rohdea japonica)叶片炭疽病菌,并明确病原菌的形态学特征和致病性。【方法】基于rDNA内转录间隔区(ITS)、甘油醛-3-磷酸脱氢酶(gapdh)、几丁质合成酶(chs)、肌动蛋白(act)、微管蛋白(tub2)这5个基因片段以及菌株的形态学特征,对分离自病斑的菌株进行准确鉴定;并通过致病性测定试验,明确菌株的致病力。【结果】多基因分子系统学研究结果显示,分离自万年青的菌株与山麦冬刺盘孢(Colletotrichum liriopes)的模式菌株(CBS 119 444)聚成一支;形态学研究结果显示,镰刀形、同侧略弯的分生孢子符合山麦冬刺盘孢的形态特征。因此,分离菌株被鉴定为白蜡树刺盘孢复合种内的山麦冬刺盘孢。本研究获得的山麦冬刺盘孢菌株接种到健康叶片上能引起典型的炭疽病斑,并能够从接种病斑中再次分离到山麦冬刺盘孢菌株。【结论】结果明确了山麦冬刺盘孢是高邮市发生严重的万年青炭疽病病原菌,为该病害的科学防控打下了基础。
【Abstract】 【Objective】To identify the leaf anthracnose fungus of Rohdea japonica, which occurs commonly in Gaoyou City,and to define the morphological characteristics and pathogenicity of the pathogen. 【Methods】The new isolates were identified by combined methods of phylogeny based on ITS-gapdh-chs-act-tub2 loci, together with morphological characters. Furthermore,pathogenicity tests were conducted using the isolates. 【Results】Phylogeny based on ITS-gapdh-chs-act-tub2 loci showed that isolates from the present study formed a clade with the type strain CBS 119 444 of Colletotrichum liriopes. In addition, conidial morphology was identical to that of C. liriopes. The isolates obtained in the present study successfully infected the healthy leaves of the host R. japonica during the pathogenicity test. 【 Conclusion】 The results clarified that C. liriopes is the pathogen of R.japonica anthracnose, which occurs severely in Gaoyou City, and laid the foundation for scientific prevention and control of this disease.
