小儿咳喘灵有效成分的定性鉴别和含量测定方法研究
Research on qualitative identification and content determination of effective components in Xiaoer Kechuanling
【摘要】 目的:建立小儿咳喘灵颗粒的薄层鉴别与含量测定的方法,加强小儿咳喘灵颗粒的质量控制。方法:采用薄层色谱法鉴别甘草、金银花、板蓝根和瓜蒌;用高效液相色谱法测定小儿咳喘灵颗粒中木犀草苷的含量,色谱柱为YMC-Pack ODS-A-C18色谱柱(250 mm x 4.6 mm,5μm),流动相为乙腈和0.5%冰醋酸溶液,梯度洗脱,检测波长为350nm。结果:薄层色谱鉴别方法专属性强,斑点清晰且阴性无干扰;木犀草苷在0.95-19μg·ml-1范围内线性关系良好,小儿咳喘灵颗粒中木犀草苷的含量为4.741μg·g-1。结论:研究建立的TLC和HPLC均简便易行,准确可靠、重现性好,可以用于小儿咳喘灵颗粒的质量控制。
【Abstract】 Objective:The method of TLC identification and content deter mination of Xiaoer Kechuanling granules was established to strengthen the quality control of Xiaoer Kechuanling granules.Method:Radix Glycyrrhizae,Flos Lonicerae,Fructus Trichosanthis and Radix Isatidis were identified by TLC.The content of luteolin in Xiaoer Kechuanling Granules was detected by HPLC.The chromatographic column was YMC-Pack ODS-A-C18 (250 mm x 4.6 mm,5μm) column.The mobile phase was acetonitrile and 0.5% glacial acetic acid solution,and the detection wavelength was 350 nm.Result:TLC identification method had strong specificity,clear spots and negative without interference.The linear range of luteolin was 0.95-~19μg·mL-1,and the content of luteolin in Xiaoer Kechuanling Granule was 4.741μg·g-1.Conclusion:The established TLC and HPLC are simple,accurate,reliable and reproducible,and can be used for the quality control of Xiaoer Kechuanling Granules.
