肽酰精氨酸脱氨酶4和组蛋白H3瓜氨酸化与高级别脑胶质瘤的相关性研究

【摘要】 目的:研究肽酰精氨酸脱氨酶4和组蛋白H3瓜氨酸化的表达与脑胶质瘤分级的关系。方法:选取2020年1月—2022年1月在河南大学淮河医院进行手术治疗的脑胶质瘤患者30例为脑胶质瘤组,其中18例为Ⅲ级、12例为Ⅳ级,取手术计划切除范围内正常脑组织为对照组。免疫组织化学检测肽酰精氨酸脱氨酶4和组蛋白H3瓜氨酸化的表达情况,并分析其与脑胶质瘤分级的关系。结果:与正常组织相比较,Ⅲ级和Ⅳ级胶质瘤组织,肽酰精氨酸脱氨酶4和组蛋白H3瓜氨酸化的表达水平上调,且上调程度与胶质瘤的级别成正比。结论:肽酰精氨酸脱氨酶4和组蛋白H3瓜氨酸化在胶质瘤组织高表达,且表达水平与级别相关,提示PAD4介导的组蛋白瓜氨酸化参与脑胶质瘤的发展。
【Abstract】 Objective: To study the relationship between the expression of peptidyl arginine deaminase 4 and histone H3 citrullination and high-grade glioma. Methods: The 30 glioma patients who underwent surgical treatment in Huaihe Hospital of Henan University were selected as the glioma group from January 2021to January 2022, of which 18 cases were grade Ⅲ and 12 cases were grade Ⅳ, and the normal brain tissue within the surgical plan were taken as the control group. The expressions of peptidyl arginine deaminase 4 and histone H3 citrullination were detected by immunohistochemistry, and their relationship with the grade of glioma was analyzed. Results: Compared with the normal tissues, the expression levels of peptidyl arginine deaminase 4 and histone H3 citrullination were up-regulated in grade Ⅲ and grade Ⅳ glioma tissues, and the up-regulation degree was proportional to the grade of glioma. Conclusion: Peptidyl arginine deaminase 4 and histone H3 citrullination are highly expression levels in glioma tissues, and the expression levels are correlated with grades, suggesting that PAD4-mediated histone citrullination is involved in the development of glioma.
