淫羊藿叶多糖提取纯化及其对小鼠酒精性肝损伤的预防作用

Extraction and Purification of Epimedium koreanum Nakai Leaf Poly-saccharide and Its Preventive Effect on Alcoholic Liver Injury in Mice

【摘要】 为探究淫羊藿叶多糖对小鼠酒精性肝损伤的保护作用及其机制,采用超声水提法提取淫羊藿叶多糖,并利用正交试验优化多糖的提取条件,然后选取纯系昆明小鼠200只,雌雄各半,随机分为空白组、模型组以及淫羊藿叶多糖低、中、高(10,20,40 mg/kg)5组,淫羊藿干预组分别提前给予30 d和2 h淫羊藿叶多糖处理,除空白组外一次性给予体积分数为56%的乙醇建立小鼠急性酒精性肝损伤模型,2 h后将小鼠处死。测定给予30 d淫羊藿叶多糖各组小鼠血清、肝脏中的天门冬氨酸基转移酶(AST)、丙氨酸基转移酶(ALT)、甘油三酯(TG)、丙二醛(MDA)、还原性谷胱甘肽(GSH)、氢氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)含量的变化,并结合病理切片考察其对小鼠酒精性肝损伤的保护效果。同时,测定提前2 h给予淫羊藿叶多糖各剂量组肝脏在5,10,15,20 h时线粒体各项指标,考察线粒体抗氧化体系功能。结果表明:所得淫羊藿叶多糖得率为5.98%,纯化后的多糖质量分数可达69.1%;淫羊藿叶多糖可显著提高小鼠血液、肝脏和肝线粒体中SOD、GSH水平,显著降低小鼠血液和肝脏以及肝线粒体中AST、ALT、MDA、TG的含量;结合病理切片发现各淫羊藿叶多糖组随剂量递增能够显著预防小鼠肝脏损伤。以上研究结果表明淫羊藿叶多糖对小鼠酒精性肝损伤具有良好的保护作用。

【Abstract】 This study optimized the extraction process conditions of Epimedium koreanum Nakai leaf polysaccharide, and explored its protective effect on alcoholic liver injury in mice and its mechanism. Firstly, ultrasonic polysaccharide extraction was used to extract polysaccharides from E. koreanum, and orthogonal conditions were used to optimize polysaccharide extraction conditions. Secondly, 200 pure Kunming mice, hale male and half female, were selected and randomly divided into blank group, model group, and low, medium and high(10, 20, 40 mg/kg) E. koreanum leaf polysaccharides groups. E. koreanum intervention group was treated with E. koreanum leaf polysaccharide for 30 d and 2 h in advance; Except for the blank group, a mouse model of acute alcoholic liver injury was established by one administration of 56% alcohol, and the mice died suddenly 2 h later. The content changes of aspartate transferase(AST), alanine transferase(ALT), triglyceride(TG), and malondialdehyde(MDA), reduced glutathione(GSH), hydroxide dismutase(SOD), and glutathione peroxidase(GPx) in serum and liver of mice in each group were measured after 30 d of administration of E. koreanum leaf polysaccharide, and the protective effect on alcoholic liver injury in mice was investigated in combination with pathological sections. At the same time, the mitochondrial indexes of liver in different doses of E. koreanum leaf polysaccharide given 2 h in advance at different time(5, 10, 15, 20 h) were measured to investigate the function of mitochondrial antioxidant system. The results showed that the yield of polysaccharides from E. koreanum leaves was 5.98%, and the content of purified polysaccharides could reach 69.1%. The results of liver injury study showed that E. koreanum leaf polysaccharides could significantly increase SOD and GSH levels significantly,and significantly reduced the contents of AST, ALT, MDA, and TG in the blood and liver and liver mitochondria of mice; combined with pathological sections, it was found that the polysaccharide groups of E. koreanum could significantly prevent liver injury in mice with increasing doses. The above results show that E. koreanum polysaccharide has a good protective effect on alcoholic liver injury in mice.
