【摘要】 目的:探讨循环肿瘤细胞(CTC) lncRNA NKILA在结肠癌放疗敏感性鉴别中的应用效果。方法:采集结肠癌放疗敏感患者和放疗耐受患者外周血CTC进行转录组测序和RT-PCR实验,分析两组患者CTC中的差异lncRNA。体外培养放疗耐受结肠癌细胞,通过细胞转染过表达细胞内NKILA,随后给予放射治疗,通过FDA/PI染色、细胞迁移实验观察放疗后细胞凋亡情况。结果:结肠癌放疗敏感患者和放疗耐受患者的CTC内lncRNA表达存在显著差异,与放疗耐受患者比较,敏感患者有9个lncRNA高表达,14个lncRNA低表达,差异有统计学意义(P<0.05),其中NKILA在敏感患者中高表达最为显著。过表达细胞内NKILA后放疗耐受结肠癌细胞对放疗的敏感性明显升高,差异有统计学意义(P<0.05)。结论:结肠癌放疗敏感患者和放疗耐受患者CTC的lncRNA表达存在显著差异,NKILA高表达与结肠癌细胞对放疗的敏感性呈正相关。更多还原

【Abstract】 Objective To investigate the application of IncRNA NKILA from circulating tumor cells(CTC) in the identification of radiation sensitivity of colon cancer.Methods Peripheral blood CTC of patients with colon cancer who were sensitive to radiotherapy and patients who were tolerant to radiotherapy were collected for transcriptome sequencing and RT-PCR to analyze the difference lncRNA in CTC of the two groups.In vitro culture of radiotherapy tolerant colon cancer cells,the intracellular NKILA was overexpressed by cell transfection.Then cell radiotherapy was given,and apoptosis was observed by FDA/PI staining and cell migration assay.Results There were significant differences in the expression of lncRNA in CTC between radiotherapy sensitive patients and radiotherapy tolerant patients.Compared with radiotherapy tolerant patients,9 lncRNAs in sensitive patients had high expression and 14 lncRNAs had low expression(P <0.05),among which NKILA had the most significant high expression in sensitive patients.The sensitivity of colon cancer cells to radiotherapy was significantly increased after overexpression of NKILA(P <0.05).Conclusion There are significant diferences in the expression of lncRNA in CTC between radiotherapy sensitive patients and radiotherapy tolerant patients of colon cancer,and high expression of NKILA is positively correlated with the sensitivity of colon cancer cells to radiotherapy.更多还原
