仔貉腹泻性奇异变形杆菌分离鉴定、致病性及耐药性检测

【摘要】 为明确引起仔貉腹泻的病原菌并分析其特征,本研究于2017年-2020年采集秦皇岛、唐山、石家庄、沧州地区患腹泻病仔貉的肛拭子、粪便以及死亡的仔貉肝脏病料组织283份,采用常规细菌分离鉴定和PCR方法对采集的病料组织进行奇异变形杆菌(Proteus mirabilis, PM)分离鉴定,采用人工感染小鼠试验、PCR方法和K-B药敏纸片法分别检测奇异变形杆菌致病性、毒力基因、耐药性及耐药基因。结果显示,分离得到109株P.mirabilis,其中78株P.mirabilis对小鼠具有不同致病性,其死亡率在40%~100%之间,毒力基因ureC、zapA、mrpA、ucaA、rsbA、pmfA、atfA检出率在79.5%~100%之间,其它毒力基因的检出率在43.6%~47.4%之间;78株致病性P.mirabilis对阿莫西林、氨苄西林、新霉素等9种药物耐药率在41.0%以上,对其它药物的耐药率在5.1%~30.8%之间,以耐10、11、12种药物的分离菌株为主,耐药基因Sul1、Sul2、adA、aac(6’)-Ib、TetA、TetM检出率在48.7%~79.50%之间,其它耐药基因检出率在2.6%~16.7%之间,耐药表型与耐药基因型之间具有一定的相关性。本研究首次报道致仔貉腹泻的奇异变形杆菌的致病性、毒力基因、耐药性及耐药基因等生物特性,为今后仔貉腹泻性奇异变形杆菌的致病机制研究、流行病学调查及防控研究提供科学依据。

【Abstract】 To identify the pathogen causing diarrhea of raccoon dogs and analyze its characteristics, 283 samples of anal swabs, feces and dead liver of young raccoon dogs suffering from diarrhea in Qinhuangdao, Tangshan, Shijiazhuang and Cangzhou from 2017 to 2020 were collected. Proteus mirabilis(PM) in these samples was identified by routine bacterial isolation and PCR. The artificially infected mice, PCR and K-B drug sensitive paper disk method were adopted to test the pathogenicity, virulence genes, drug resistance and drug resistance genes of Proteus mirabilis. The results showed that 109 strains of PM were isolated, of which 78 strains had different pathogenicity to mice, with the mortality being 40%~100%. The detection rates of virulence genes ureC, zapA, mrpA, ucaA, rsbA, pmfA and atfA were 79.5%~100%, and the detection rates of other virulence genes were 43.6%~47.4%. The resistance rates of 78 pathogenic PM strains to amoxicillin, ampicillin and neomycin were more than 41.0%, and the resistance rates to other drugs were 5.1% ~ 30.8%. The isolates resistant to 10, 11 and 12 drugs were dominant. The detection rates of resistance genes Sul1, Sul2, adA, aac(6’)-Ib, TetA and TetM were between 48.7% and 79.50%, and the detection rates of other resistance genes were from 2.6% to 16.7%, and there was a correlation between drug resistance phenotype and drug resistance genotype. In this study, the pathogenicity, virulence genes, drug resistance and drug resistance genes of Proteus mirabilis causing diarrhea in raccoon dogs are reported for the first time, so that it provides a scientific basis for the pathogenic mechanism, epidemiological investigation and prevention and control of Proteus mirabilis causing diarrhea in raccoon dogs in the future.
