抗过氧化氢致人皮肤角质形成细胞氧化损伤活性物质的筛选及其机制初探
Screening of compound of anti-oxidative damage of HACAT cells induced by H2O2 and preliminary study on its mechanism
【摘要】 目的 筛选具有保护人皮肤角质形成细胞(HACAT)抗过氧化氢(H2O2)氧化损伤的活性样品,并探索其抗氧化的机制。方法 用CCK-8法测定不同植物样品对H2O2损伤HACAT细胞的保护作用,获得活性样品远志寡精F。检测远志寡精F对损伤细胞的增殖活性、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)的活性、丙二醛(MDA)、活性氧(ROS)、白细胞介素(IL)-6、IL-8的含量以及细胞凋亡和坏死的影响。结果 经筛选得到的活性物质远志寡精F能提高损伤细胞的增殖活力、SOD及GSH-Px活性,减少MDA、ROS的产生和IL-6、IL-8的表达,降低细胞的凋亡率和坏死率。结论 远志寡精F可能是通过增加抗氧化活性物质的产生、减轻炎症反应、减少细胞凋亡和坏死来发挥抗氧化损伤的作用。
【Abstract】 OBJECTIVE To screen the active samples which can protect HACAT cell against H2O2 oxidative damage, and to explore its mechanism.METHODS The protective effects of different plant samples on H2O2 damaged HACAT cells were determined by CCK-8 to obtain the active samples.The effect of active samples on proliferation, activities of SOD and GSH-Px, contents of MDA,ROS,IL-6 and IL-8,apoptosis and necrosis of injured cells were detected.RESULTS Tenuifolioses F was obtained by screening.It can increase the proliferation, activities of SOD and GSH-Px, decrease the levels of MDA,ROS,IL-6 and IL-8,reduce the apoptosis and necrosis of injured HACAT cells.CONClUSION Tenuifolioses F may play an anti-oxidative injury role by increasing antioxidant active substances, reducing inflammatory response, and reducing cell apoptosis and necrosis.
