连翘叶内源酶转化木脂素苷产生木脂素苷元工艺研究
Study on lignan aglycones production from lignan glycosides transformed by endogenous enzymes in Forsythia suspensa leaves
【摘要】 探究连翘叶木脂素苷类成分经生物转化产生木脂素苷元的较优发酵条件。利用HPLC法建立发酵叶中连翘脂素、松脂素、表松脂素、松脂素单甲基醚含量的测定方法;考察连翘叶内源粗酶液对木脂素苷的体外转化作用;在单因素实验的基础上,以木脂素苷元含量为指标,采用正交试验对连翘叶发酵转化工艺进行优化。结果表明连翘叶中存在水解木脂素苷类的酶;较优的工艺参数为发酵温度55℃,发酵湿度55%,发酵时间50 min。发酵后四种木脂素苷元总含量从0.33%增加至5.66%,约提高了17倍。该工艺条件可有效提高连翘叶中连翘脂素、松脂素、表松脂素、松脂素单甲基醚含量,为进一步从连翘叶中制备四种木脂素苷元提供了理论依据。
【Abstract】 The optimal fermentation conditions for the production of lignan aglycones using the biotransformation of lignan glycosides in Forsythia suspensa leaves was investigated.HPLC method was established to measure the concentrations of phillygenin, pinoresinol, epipinoresinol, and pinoresinol monomethyl ether in fermented leaves and to investigate the in vitro transformation of lignan glycosides by endogenous crude enzyme solution.The orthogonal optimization method was used to select the process on the basis of a single factor experiment using the number of lignan aglycones as the index.The optimum process was a fermentation temperature of 55℃,fermentation humidity 55%,and fermentation time of 50 min.The lignan aglycone content increased approximately 17-fold from 0.33%to 5.66%after fermentation.This procedure had the potential to increase the concentrations of phillygenin, pinoresinol, epipinoresinol, and pinoresinol monomethyl ether and offered a theoretical framework for further lignan aglycone preparation.
