益气、活血、化痰法不同组合对COPD大鼠炎症反应及气道黏液高分泌的干预作用
The Intervention of Different Combinations of Yiqi (益气), Huoxue (活血) and Huatan (化痰) Treatment on the Inflammatory Reaction and Mucosal Hypersecretion of Airway Mucus in COPD Rats
【摘要】 目的:观察益气、活血、化痰法不同治疗方案对慢性阻塞性肺疾病(COPD)模型大鼠血清中IL-8、TNF-α及肺泡灌洗液(BALF)中水通道蛋白5(AQP5)、黏蛋白5AC(MUC5AC)的调节作用。方法:54只SD雄性大鼠按随机数字表法分为空白组、模型组、益气化痰组、益气活血组、活血化痰组及益气活血化痰组,每组9只。其中空白组不造模,其余各组大鼠采用香烟烟熏结合LPS气管滴注法建立COPD模型。从造模当天进行灌胃治疗,1次/d,28 d后处死大鼠。取血清、肺泡灌洗液(BALF)及左侧肺组织,苏木精-伊红染色(HE)观察肺组织病理,酶联免疫吸附法(ELISA)测定大鼠BALF中AQP5、MUC5AC及血清中IL-8、TNF-α的含量。结果:与模型组比较,益气活血组、益气化痰组、活血化痰组及益气活血化痰组大鼠肺组织和气道结构较完整,炎症细胞浸润程度较轻,各组中药干预后均可一定程度上改善COPD模型大鼠的肺组织病理损害。与空白组比较,模型组大鼠血清中IL-8、TNF-α含量均明显升高(P<0.05);与模型组比较,益气活血组、益气化痰组、活血化痰组及益气活血化痰组大鼠血清中IL-8、TNF-α含量均明显降低(P<0.05),且各中药干预组间大鼠血清中TNF-α含量比较,差异无统计学意义(P>0.05)。与空白组比较,模型组大鼠BALF中AQP5含量明显降低(P<0.05),MUC5AC含量明显升高(P<0.05);与模型组比较,益气活血组、益气化痰组、活血化痰组及益气活血化痰组大鼠BALF中AQP5含量均明显升高(P<0.05),MUC5AC含量均明显降低(P<0.05)。结论:益气、活血、化痰法不同组合治疗方案对COPD的疗效可能与抑制炎症反应和缓解气道黏液高分泌密切相关。
【Abstract】 Objective: To observe the effects of different treatment regimens of Yiqi, Huoxue and Huatan on t he levels of inflammatory cytokines IL-8 and TNF-α, as well as the moderation effect on aquaporin5(AQP5) and mucin5AC(MUC5AC) of alveolar lavage fluid(BALF) in rats with chronic obstructive pulmonary disease(COPD). Methods: Totally 54 SD rats were randomly divided into blank group, model group, Yiqi Huatan group,Yiqi Huoxue group, Huoxue Huatan group and Yiqi Huoxue Huatan group, 9 rats in each group. Except for the blank group, COPD rats were established by tracheal instillation of cigarette smoke and lipopolysaccharide in the other groups. The rats were killed after 28 days on the day of modeling, and the serum, BALF and left lung tissue were collected. Lung tissue pathology was observed according to histopathology(hematoxylin-eosin) staining.And enzyme-linked immunosorbent assay(ELISA) was used to detect the levels of AQP5 and MUC5AC in BALF and serum IL-8 and TNF-α. Results: Compared with the model group, the lung tissue and airway structure of rats were relatively intact, and the degree of inflammation infiltration was milder in Yiqi Huoxue group, Yiqi Huatan group, Huoxue Huatan group and Yiqi Huoxue Huatan group. After Chinese medicine intervention, the lung tissue pathological damage in COPD model rats was improved to a certain extent. Compared with the blank group, the levels of serum IL-8 and TNF-α were significantly increased in model group(P <0.05). Compared with the model group, the levels of serum IL-8 and TNF-αwere significantly decreased in Yiqi Huoxue group, Yiqi Huatan group, Huoxue Huatan group and Yiqi Huoxue Huatan group(P<0.05). There was no significant difference in TNF-α levels among the Chinese medicine intervention groups(P>0.05). Compared with the blank group, the AQP5 level in BALF was significantly decreased in model group(P<0.05), while the MUC5AC level was significantly increased(P<0.05). Compared with the model group, the levels of AQP5 in BALF were significantly increased in Yiqi Huoxue group, Yiqi Huatan group, Huoxue Huatan group and Yiqi Huoxue Huatan group(P<0.05), and the levels of MUC5AC were significantly decreased(P<0.05). Conclusion: The curative effect of different combination therapy of Yiqi, Huoxue and Huatan on COPD may be closely related to inhibiting inflammation reaction and relieving airway mucus hypersecretion.
