荣脉软胶囊改善缺铁性贫血的动物实验研究

【摘要】 目的 观察荣脉软胶囊对实验性大鼠改善缺铁性贫血的作用。方法 选用刚断乳雌性SD大鼠80只,用低铁饲料喂养,剪尾放血构建低铁性贫血大鼠模型,按血红蛋白(Hb)水平随机分为低铁对照组(色拉油)、阳性对照组(硫酸亚铁)和3个受试物组,受试物组(低、中、高剂量组,分别为0.2 g/kg、0.4 g/kg和1.2 g/kg),每组10只。每天经口灌胃1次,连续30 d。试验结束测定大鼠Hb、红细胞内游离原卟啉含量和红细胞压积。结果 试验后中、高剂量组大鼠Hb含量分别为(103.0±12.4) g/L、(111.0±6.3) g/L,与低铁对照组(86.6±9.6) g/L相比明显升高(P均<0.05);红细胞压积分别为(37.9±3.8)%、(39.8±2.5)%,与低铁对照组(33.1±3.3)%相比明显升高(P<0.05)。高剂量组大鼠红细胞内游离原卟啉含量为(320.6±72.6)μg/L,与低铁对照组(437.0±93.2)μg/L相比明显降低(P<0.05)。结论 荣脉软胶囊对实验性大鼠的缺铁性贫血具有改善作用。

【Abstract】 Objective To observe the effect of rongmai soft capsule on iron deficiency anemia in experimental rats. Methods A total of 80 newly weaned female SD rats were selected, and the rat model of low iron anemia was established by feeding with low iron diet, and tail cutting and blood letting. According to the level of Hb, they were randomly divided into low iron control group(salad oil), positive control group(ferrous sulfate) and three test substance groups(0.2 g/kg, 0.4 g/kg and 1.2 g/kg), with 10 rats in each group. The test substance was administered by oral gavage once a day, for 30 consecutive days. At the end of the experiment, Hb content, free protoporphyrin(FEP) content in erythrocytes and hematocrit were measured. Results The Hb content of rats in the middle and high dose groups were(103.0±12.4) and(111.0±6.3) g/L, which was significantly higher than that in the low iron control group(86.6±9.6) g/L(all P<0.05) after the test; the hematocrit was(37.9±3.8)% in the middle dose group and(39.8±2.5)% in the high dose group, respectively, which was significantly higher than that in the lowiron control group(33.1±3.3)%(P<0.05). The content of free protoporphyrin in the erythrocytes of rats in the high dose group was(320.6±72.6) μg/L, which was significantly lower than that in the lowiron control group(437.0±93.2) μg/L(P<0.05). Conclusion Rongmai soft capsule can improve iron deficiency anemia in experimental rats.
