嗜冷假单胞菌菌株F3的筛选及其中低温脱氮特性

【摘要】 从内蒙古某生活污水处理厂的活性污泥中筛选出1株耐低温高效脱氮菌株F3,综合使用16S r RNA基因序列分析及形态、生理生化鉴定方法对其系统发育地位进行了初步鉴定,并探究了环境因子对菌株影响,评测了菌株在5~25℃下的脱氮性能.菌株F3的革兰氏染色为阴性、无芽孢,与嗜冷假单胞菌(Pseudomonas psychrophila)的16S rRNA基因序列一致性最高.在15℃温度下,菌株F3在盐度为0~5 g/L、中性(pH 7)、碳氮比为23的条件下生长代谢能力最佳,其对化学需氧量(COD)和氨氮(NH4+-N)去除率可分别达91%和86%.菌株F3在5~25℃下具备较好的异养硝化-好氧反硝化脱氮能力.

【Abstract】 A psychrotolerant bacterial strain F3 with efficient nitrogen-removal capability was screened from the activated sludge of a domestic sewage treatment plant in Inner Mongolia.Combining morphological,physiological and biochemical identification methods with 16S rRNA gene sequence analysis,we determined the phylogeny of F3.Additionally,the effects of environmental factors on the growth of F3 were explored,and the nitrogen-removal performance at 5-25℃was evaluated.Strain F3 was Gram-negative and non-spore-forming,sharing the highest16S r RNA gene sequence identity with Pseudomonas psychrophila.At 15℃,salinity of 0-5 g/L(Na Cl concentration),carbon to nitrogen ratio of 23,and pH 7,strain F3 grows well and could remove 91%of COD and 86%of ammonia nitrogen.Strain F3 showed efficient performance of heterotrophic nitrification aerobic denitrification at 5-25℃.
