桑叶水提工艺优化及水提物的体外抑菌试验
Optimization of the Aqueous Extraction Process of Morus Folium and the in Vitro Antibacterial test of the Aqueous Extracts
【摘要】 为优化桑叶水提工艺并研究其水提物的体外抗菌效果,首先采用单因子试验,分别探索提取次数、时间和溶剂倍数对桑叶水提浸膏得率的影响;然后根据单因子试验的结果优选出相应的工艺参数进行正交试验,以浸膏得率与总黄酮含量为考察指标,筛选出最佳的提取工艺组合;最后,采用纸片法研究桑叶水提物的体外抑菌作用。结果表明:(1)在提取次数3次、提取时间1 h、溶剂倍数18∶1 mL/g的工艺条件下,桑叶水提的平均浸膏得率最高,达31.13%,总黄酮平均含量也最高,达32.12 mg/g;(2)桑叶水提物对金黄色葡萄球菌、大肠杆菌、农杆菌的抑菌圈直径分别为16.14±1.96、8.31±0.21、9.76±0.72(mm),而在沙门氏菌、铜绿假单胞菌、枯草芽孢杆菌中未出现抑菌圈。结果显示:桑叶水提的工艺条件以提取次数3次、提取时间1 h、溶剂倍数18∶1 mL/g为最佳;桑叶水提物对金黄色葡萄球菌、大肠杆菌、农杆菌有一定的抑制作用,而对其余三种菌无抑菌作用。
【Abstract】 In order to optimize the extraction process and explore the in vitro antibacterial effect of Morus folium aqueous extract, firstly, a one-factor test was conducted to explore the effects of the extractions times, extraction time, and solvent ratio on the extraction rate of Morus folium, respectively; then, according to the results of the one-factor test, the corresponding process parameters were selected for an orthogonal test, in which the extraction rate and the total flavonoid content in the extract were used as the indexes for investigation to select the optimal combination of extraction processes parameters; finally, a paper slide method was used to explore the in vitro antibacterial effects of Morus folium aqueous extract. The results showed that:(1)Under the process conditions of 3 times of extraction, 1 h of extraction time, and 18∶1 mL/g of solvent ratio, the aqueous extract of Morus folium showed the highest average extraction rate of 31.13% and the highest average total flavonoid content of 32.12 mg/g.(2)The diameters of the inhibition circles of Morus folium aqueous extract to Staphylococcus aureus, Escherichia coli and Agrobacterium were 16.14±1.96, 8.31±0.21, 9.76±0.72(mm)respectively, and no inhibition circle appeared in Salmonella, Pseudomonas aeruginosa, and Bacillus subtilis. It prompted that the optimal process parameters for the aqueous extract of Morus folium were 3 times of extraction, 1 h of extraction time, and 18:1 mL/g of solvent ratio; the aqueous extract of Morus folium have a certain inhibitory effect on Staphylococcus aureus, Escherichia coli, and Agrobacterium, with no inhibitory effects on the remaining three bacteria.
