乾华肉用美利奴羊精液稀释保存的研究与应用效果评价

【摘要】 为了研究常温和低温环境下不同稀释液对乾华肉用美利奴羊精液保存效果的影响,试验采集6只乾华肉用美利奴羊种公羊精液,取合格精液混匀后均分为7份,其中4份利用A液(萄萄糖-柠檬酸钠)、B液(萄萄糖-乳糖-柠檬酸钠)、C液(萄萄糖-蔗糖-柠檬酸钠)、D液(萄萄糖-果糖-柠檬酸钠-EDTA)稀释后进行常温(15~18℃)保存,3份利用E液(萄萄糖-柠檬酸钠-卵黄)、F液(果糖-柠檬酸钠-卵黄)和G液(生理盐水-卵黄)稀释后进行低温(2~4℃)保存,均保存24 h后测定精子体外存活时间和存活指数,选择最适常温和低温保存稀释液;然后将720只同期发情的乾华肉用美利奴羊种母羊随机分为6组,每组120只,利用上述筛选的常温和低温最适稀释液保存的0 h(精液稀释后立即输精)及6,12 h乾华肉用美利奴羊精液进行人工授精,测定母羊繁殖性能指标(第一情期妊娠母羊数、总妊娠母羊数、产羔数、第一情期受胎率、总受胎率、产羔率),筛选最适乾华肉用美利奴羊精液的保存方式。结果表明:常温条件下,C液保存的精子体外存活时间和存活指数均显著优于A液、B液、D液(P<0.05);低温条件下,F液保存的精子体外存活时间和存活指数显著高于E液、G液(P<0.05);利用C液进行常温保存的精液的平均第一情期受胎率、总受胎率和产羔数均显著高于利用F液进行低温保存的精液(P<0.05)。说明最适乾华肉用美利奴羊精液保存条件为利用C液在常温下保存。

【Abstract】 In order to study the effects of different diluents on the semen preservation of Qianhua mutton Merino Sheep at room temperature and low temperature, semen samples from six Qianhua mutton Merino Sheep were collected, and the qualified semen were mixed and divided into seven parts. Among them, four parts were diluted with solution A(glucose-citrate sodium), solution B(glucose-lactose-citrate sodium), solution C(glucose-sucrose-citrate sodium), and solution D(glucose-fructose-citrate sodium-EDTA) and then preserved at room temperature(15-18 ℃), 3 parts were diluted with solution E(glucose-citrate sodium-yolk), solution F(fructose-citrate sodium-yolk), and solution G(physiological saline-yolk) and then preserved at low temperature(2-4 ℃). The survival time and survival index of spermatozoa in vitro were measured after 24 h of storage, and the optimal diluent for room temperature and low temperature preservation was selected. Then 720 Qianhua mutton Merino ewes in estrus at the same time were randomly divided into six groups. There were 120 sheep in each group. Artificial insemination was performed with the semen of Qianhua mutton Merino sheep stored in the optimal diluent at room temperature and low temperature for 0(semen was immediately inseminated after dilution), 6 and 12 h. The reproductive performance indexes of ewes(number of pregnant ewes in the first estrus, total number of pregnant ewes, number of lambs, conception rate in the first estrus, total conception rate, lambing rate) were measured to screen the best way to preserve semen of Qianhua mutton Merino sheep. The results showed that the survival time and survival index of spermatozoa stored in solution C at room temperature were significantly better than those in solution A, B and D(P<0.05). The survival time and survival index of sperm in diluent F were significantly higher than those in solution E and G(P<0.05). The average first estrus pregnancy rate, total pregnancy rate and litter number in solution C stored at room temperature were significantly higher than those in solution F stored at low temperature(P<0.05). The results indicated that the optimal preservation condition of semen of Qianhua mutton Merino sheep was solution C stored at room temperature.
