响应面法优化石榴皮总黄酮提取工艺及其抗氧化性能研究
Optimization in Extraction Process of Total Flavonoids from Pomegranate Peel by Response Surface Methodology and Its Antioxidant Property
【摘要】 在单因素实验的基础上,采用响应面法对石榴皮总黄酮的醇提工艺进行了优化,并对石榴皮总黄酮的抗氧化性能进行了评价。结果表明,石榴皮总黄酮最优醇提工艺条件如下:乙醇体积分数70%、料液比1∶21(g∶mL)、提取温度80.5℃、提取时间3.0 h,在此条件下,石榴皮总黄酮提取量达到58.87 mg·g-1,与理论预测值(59.801 1 mg·g-1)的相对误差为1.58%。提取得到的石榴皮总黄酮具有一定的抗氧化活性,对DPPH自由基的清除率随浓度增加逐渐升高。
【Abstract】 On the basis of single-factor experiments, we optimized the ethanol extraction process of total flavonoids from pomegranate peel by response surface methodologies, and evaluated the antioxidant property of total flavonoids from pomegranate peel.The results show that the optimal ethanol extraction conditions for total flavonoids from pomegranate peel are determined as follows: the ethanol volume fraction of 70%,the solid-liquid ratio of 1∶21(g∶mL),the extraction temperature of 80.5 ℃,and the extraction time of 3.0 h.Under above conditions, the extraction amount of total flavonoids from pomegranate peel can reach 58.87 mg·g-1,with a relative error of 1.58% compared to the theoretical prediction value(59.801 1 mg·g-1).The total flavonoids extracted from pomegranate peel have certain antioxidant activity, and the scavenging rate of DPPH free radicals gradually increases with the increase of concentration.
