【摘要】 本实验用胶体金标记RAC单克隆抗体构建一种能够快速检测RAC的免疫层析试纸条，通过正交试验优化试纸条中标记pH、抗体的量、NC膜上RAC抗原及羊抗鼠抗体划线的浓度等因素得到最佳组合，并测定试纸条的特异性、稳定性、检测时间和检测限。结果表明：经过优化后得到的最佳组合为pH=6,标记抗体的量4μg, RAC抗原及羊抗鼠抗体的浓度分别为0.8 mg/mL和1.0 mg/mL;该试纸条可在5 min内特异性地检测RAC,检测限为2 ng/mL,在室温条件下能保存12个月。更多还原

【Abstract】 The colloidal gold marked RAC monoclonal antibody was used to build quick detection of RAC immune chromatography test paper, and pH, amount of antibodies, the concentrations of RAC antigen and goat antimouse antibody was optimized through the orthogonal experiment, and the specificity, stability and detection limit of the test strips was determined. Experimental results showed that the best preparation Canditions were as follows: pH=6, RAC antibody was 4 μg/mL, RAC antigen was 0.8 mg/mL, and goat antimouse antibody was 1.0 mg/mL. The best preparation conditions were as follows: The test strip could detect RAC within 5 min with detection limit 2 ng/mL, and could be stored for 12 months.更多还原
