细柱西番莲高效再生体系的建立
Establishment of an Efficient Regeneration System in Passiflora suberosa L.
【摘要】 [目的]建立细柱西番莲高效再生体系,为细柱西番莲诱变育种和分子育种提供依据。[方法]以细柱西番莲真叶、叶柄和带芽茎段为外植体,研究植物生长调节剂对愈伤组织诱导、不定芽诱导、丛生芽的增殖和壮苗生根的影响。[结果]细柱西番莲的真叶和叶柄在添加不同浓度6-BA的MS培养基上均能诱导产生愈伤组织,真叶在1.0 mg/L6-BA的MS培养基上先诱导愈伤组织后形成不定芽,不定芽诱导率为51.67%,增殖系数达到5.1;带芽茎段在1.0 mg/L6-BA的MS培养基上能直接产生不定芽,增殖系数达到5.99;MS+1.0 g/L活性碳+1.0 mg/L IBA培养基适合小芽壮苗生根。组培苗移栽于泥炭土中,成活率可达83.33%。[结论]单独添加6-BA能较好地诱导细柱西番莲愈伤组织和不定芽的再生与增殖。
【Abstract】 [Objective] The purpose of was to establish an efficient regeneration system for passionflower,providing support for mutation and molecular breeding of Passiflora suberosa L. [Method] The effects of plant growth regulators on callus induction,adventitious bud induction,proliferation of clustered buds,and rooting of strong seedlings were studied using the leaves,petioles,and sprouted stem segments of passionflower as explants. [Result] The leaves and petioles of P. suberosa could induce callus formation on MS medium supplemented with different concentrations of 6-BA. On MS medium containing 1.0 mg/L 6-BA,callus tissue could be induced from leaves and then form adventitious buds. The induction rate of adventitious buds was51.67%,and the proliferation coefficient was 5.1. The stem segments with buds could directly produce adventitious buds on MS medium containing 1.0 mg/L 6-BA,with a proliferation coefficient of 5.99. The MS medium containing 1.0 g/L activated carbon and 1.0 mg/L IBA was suitable for rooting of tissue cultured seedlings. The survival rate of regenerated seedlings transplanted into peat soil was 83.33%. [Conclusion] The use of 6-BA alone could effectively induce the regeneration and proliferation of callus and adventitious buds in P. suberosa.
