表皮生长因子样重复系列和盘状蛋白Ⅰ样结构域3对宫颈癌细胞增殖和迁移的作用研究
Study on the effect of epidermal growth factor-like repeat and discoidin Ⅰ-like domain 3 on proliferation and migration of cervical cancer cells
【摘要】 目的 探讨表皮生长因子样重复系列和盘状蛋白I样结构域3(EDIL3)在宫颈癌中的表达及其对宫颈癌细胞的增殖、迁移能力与上皮-间质转化的作用。方法 将SiHa、HeLa细胞分为2组:对照组(转染空质粒)和实验组(转染过表达EDIL3质粒)。用噻唑蓝(MTT)法和细胞划痕实验分别评估宫颈癌细胞的增殖和迁移能力;用蛋白质印迹法、实时定量聚合酶链反应(qRT-PCR)测定EDIL3和上皮-间质转化相关分子在细胞中的表达水平。结果 HaCaT、SiHa、HeLa、C33A细胞中EDIL3蛋白表达水平为0.86±0.08,0.59±0.10,0.40±0.02和0.45±0.07。SiHa对照组、SiHa实验组、HeLa对照组、HeLa实验组细胞96 h的增殖活性分别为3.86±0.44,4.69±0.44,3.39±0.37和4.69±1.28;划痕愈合率分别为0.21±0.02,0.29±0.02,0.32±0.01和0.43±0.04;钙黏蛋白E表达水平分别1.09±0.13,0.64±0.17,0.87±0.03和0.62±0.11;β-连环蛋白表达水平分别为0.28±0.06,0.45±0.02,0.67±0.02和0.89±0.07;波形蛋白表达水平分别为0.27±0.03,0.43±0.07,0.43±0.03和0.54±0.05;锌指转录因子表达水平分别为0.27±0.03,0.47±0.02,0.42±0.04和0.61±0.08。上述指标,同种细胞实验组与对照组比较,差异均有统计学意义(P<0.05,P<0.01)。结论 EDIL3在宫颈癌中低表达,而过表达EDIL3可促进宫颈癌细胞的增殖、迁移和上皮-间质转化。
【Abstract】 Objective To investigate the expression of epidermal growth factor-like repeat series and discoid protein I-like domain 3(EDIL3) in cervical cancer and their effects on the proliferation and migration of cervical cancer cells as well as epithelial-mesenchymal transition. Methods The expression of EDIL3 in cervical tissues was analyzed by immunohistochemistry. The SiHa and HeLa cells were divided into two groups: the control group(transfected with empty plasmid) and the experimental group(transfected with the expression of EDIL3 plasmid). The proliferation and migration abilities of cervical cancer cells were evaluated by methyl thiazolyl tetrazolium(MTT) method and cell scratch test, respectively. The expression levels of EDIL3 and epithelial-mesenchymal transition related molecules in cells were determined by Western blot and real-time quantitative polymerase chain reaction ( qRT-PCR). Results The protein expression levels of EDIL3 in Ha Ca T,Si Ha,He La and C33A cells were 0. 86 ±0. 08,0. 59 ± 0. 10,0. 40 ± 0. 02,0. 45 ± 0. 07. The 96 h cells proliferation rates of Si Ha NC,Si Ha EDII3,He La NC and He La EDIL3 groups were 3. 86 ± 0. 44,4. 69 ± 0. 44,3. 39 ± 0. 37 and 4. 69 ± 1. 28,respectively; the scratch healing rates were 0. 21 ± 0. 02,0. 29 ± 0. 02,0. 32 ± 0. 01 and 0. 43 ± 0. 04,respectively; the E-cadherin protein expression levels were 1. 09 ± 0. 13,0. 64 ± 0. 17,0. 87 ± 0. 03 and 0. 62 ± 0. 11,respectively; the protein expression levels of β-catenin were 0. 28 ± 0. 06,0. 45 ± 0. 02,0. 67 ± 0. 02 and 0. 89 ± 0. 07,respectively; the protein expression levels of Vimentin were 0. 27 ± 0. 03,0. 43 ± 0. 07,0. 43 ± 0. 03 and 0. 54 ± 0. 05,respectively; the protein expression levels of Slug were 0. 27 ± 0. 03,0. 47 ± 0. 02,0. 42 ± 0. 04 and 0. 61 ± 0. 08,respectively. The differences of the above indexes between the experimental group and the control group in same type of cell were statistically significant( P < 0. 05,P < 0. 01). Conclusion EDIL3 is lowly expressed in cervical cancer,and overexpression of EDIL3 can promote the proliferation,migration and epithelial-mesenchymal transition of cervical cancer cells.
