巴西橡胶树乳管细胞中与HbMago2相互作用蛋白的筛选与分析

【摘要】 为探索HbMago2在乳管细胞中的作用,本研究构建了HbMago2的诱饵载体,从巴西橡胶树乳管cDNA表达文库中筛选与HbMago2相互作用的蛋白。结果显示,诱饵载体无自激活性、对酵母菌Y2HGold无毒性;通过文库筛选,获得3个与HbMago2相互作用的蛋白;点对点验证结果表明HbMago2能够与Y14、phox和HMAD在酵母中能相互作用。互作蛋白中phox是烟酰胺腺嘌呤二核苷酸磷酸氧化酶(NADPH)的亚基,NADPH氧化酶能引起ROS,而重金属相关结构域蛋白(HMAD)能清除重金属引起的ROS,推测HbMago2通过蛋白互作协调乳管细胞中ROS的平衡。本研究结果为进一步研究HbMago2在乳管细胞中的功能提供重要的研究基础。

【Abstract】 In order to explore the functions of HbMago2 in laticifer cells, a bait vector of HbMago2 was constructed for yeast two-hybrid, and proteins that interacted with HbMago2 were screened from a latex cDNA expression library of Hevea brasiliensis in this study. The results showed that the bait vector had no autoactivation and was non-toxic to the yeast strain Y2HGold. Through library screening, we obtained three proteins from a yeast two-hybrid library of laticifer cells c DNA using yeast two-hybrid system. One-to-one testing results showed that HbMago2 could interact with Y14, phox and HMAD in yeast. The phox is a subunit of nicotinamide adenine dinucleotide phosphate oxidase(NADPH) in interacting proteins. NADPH oxidase enzyme could cause ROS, and heavy-metal-associated domain-containing protein(HMAD) could clear ROS caused by heavy metals. We inferred that HbMago2 coordinates the balance of ROS in laticifer cells through protein-protein interactions. The results of this study provide an important research foundation for further investigating the function of HbMago2 in laticifer cells.
